Effect of Zhi-Tong-Xiao-Zhong Powder on Serum Level of IL-1β and TNF-α in Rabbit Knee Joint Model

Abstract:
This study was aimed to observe the effect of Zhi-Tong-Xiao-Zhong(ZTXZ) powder,which is an external application,on knee osteoarthritis rabbit model.Changes on serum level of IL-1β and TNF-α were measured to clarify the mechanism of ZTXZ powder in the treatment of rabbit knee osteoarthritis.A total of 36 rabbits were randomly divided into 6 groups.The brake method was used in all groups except the blank control group to make the rabbit knee osteoarthritis model.ZTXZ powder was used to wash on the knee joint of rabbit model externally for two weeks.Then,rabbit models were killed and the blood serum was separated from the blood sample.The enzyme-linked immunosorbent assay technique(ELSIA) was used in the detection of serum level of IL-1β and TNF-α.The results showed that compared with model group,there was significant difference in the normal control group on serum level of IL-1β and TNF-α(P<0.01).Compared to normal control group,there were no differences among treatment groups(P>0.05).The serum level of IL-1β and TNF-α in model group and other treatment groups were decreased at different levels(P<0.05).There was no difference among the low,medium and high dose of ZTXZ powder group with Gu-You-Ling liniment group(P>0.05).There was no difference on the serum level of IL-1β among low,medium and high dose of ZTXZ powder group(P>0.05),while there was significant difference on the serum level of TNF-α(P<0.05).It was concluded that ZTXZ powder can reduce the serum level of IL-1β and TNF-α in the rabbit knee osteoarthritis model.It showed that the ZTXZ powder is able to reduce the effect of inflammatory cytokines in osteoarthritis.This study has provided an objective scientific basis for further clinical application of ZTXZ powder and drug development.
