A preliminary study on conservation of Andrias davidanus semen in vitro

Abstract:
Gonadal development of Andrias davidanus not synchronized,semen characteristics and preservation conditions of A.davidanus were studied targeting the increase of fertility rate.Semina were collected by artificial assistance approach,and a regular assay was conducted on the basic features such as semen volume,pH,density,vitality,etc.Preservation conditions,namely,protection medium,illumination and temperature,were also studied.The results showed that semen volume of pro-male A.davidanus was up to 12 mL/kg,pH 6.7-7.0 of subacid condition;and that the semen density was(2.15-10.75)million/mL,sperm motility 0.7-0.9.In vitro conservation conditions,dark environment at ambient illuminance(20 lx).The result also indicated that the survival time of the semina was 0.5 h longer than those conserved in the luminous environment(400 lx);low temperature(0-4℃) could extend the survival time of the semina 3d longer than those at room temperature(21-22℃) in dark environment;in the cold dark environment,semina with protection medium added in could extend the survival time 6-12 h longer than the control group.
