INTERACTION BETWEEN AVIDIN AND BIOTIN-CONTAINING SUPPORTED PLANAR MEMBRANE

Abstract:
Two-dimensional assmble of biological molecules is one important field in field in the molecular engineering. We.studied the interaction of avidin with phospholipid model membranes con taining biotin-conjugated phospholipids. Using LB technique together with sarface sensitive techniques (such as eilipsometry and sarface plasmon spectroscopy), we studied the nonspecific adsorption and specific binding of avidin to phospholipid lamella, and the effect of the lateral steric hindrance.The primary conclusions of the present work are as following first, the charges of membrane sarface effect the rate of protein adsorption, but do not effect the amount of protein ultimately adsorbed second, the nonspecific adsorption of avidin can be detached by adding a suitable amount of bivalent alkaline-earth metal ions; third, the binding of avidin to membrane bound model receptor is astricted remarkable by the latelal steric hindrence.
