Study on the Lipid-Lowering Effect and Mechanism of Suoquan Yishen Prescription on Caenorhabditis elegans %A 邵恩茜 %A 庄燕苹 %A 张鈺明 %A 陈小凡 %A 唐昕 %A 刘洋 %A 陈柏岑 %A 肖曼 %A 杨帆 %A 谢毅强 %J Traditional Chinese Medicine %P 1257-1266 %@ 2166-6059 %D 2024 %I Hans Publishing %R 10.12677/tcm.2024.136191 %X 目的:以N2野生型秀丽隐杆线虫为研究对象,探究缩泉益肾方(SQYSP)对线虫脂质代谢的影响及作用机制。方法:SQYSP以醇提和水提两种不同方式提取,并分别以20、50、100 μg/mL的剂量作用于秀丽隐杆线虫,观察SQYSP对线虫运动能力和生殖能力的影响。采用油红O染色法对线虫肠道脂滴进行染色,观察线虫体内脂肪沉积的变化;同时,通过RT-PCR检测线虫体内与脂质代谢相关基因skn-1、sbq-1、fasn-1、fat-7的表达水平。结果:SQYSP醇提物在50 μg/mL和100 μg/mL浓度下均可以提高线虫寿命(P < 0.05, P < 0.001),SQYSP水提物仅在100 μg/mL浓度下提高线虫寿命(P < 0.01)。此外,两种提取物在100 μg/mL浓度下均可以提高线虫的身体弯曲频率(P < 0.01, P < 0.05)。油红O实验中,高浓度SQYSP醇提物和水提物均可以改善线虫体内的脂质堆积情况(P < 0.001, P < 0.001),且RT-PCR实验显示,两种提取物均可以提高skn-1和降低sbq-1、fasn-1、fat-7的表达量(P < 0.01)。结论:SQYSP的醇提物和水提物在不损害线虫正常生理功能的情况下,对秀丽隐杆线虫有降脂的作用,其作用机制可能与skn-1、sbq-1、fasn-1和fat-7的调节有关。
Objective: To study the effect of Suoquan Yishen Prescription (SQYSP) on lipid metabolism of N2 wild type Caenorhabditis elegans and its possible mechanism. Methods: SQYSP was extracted by ethanol and water, and then administered to Caenorhabditis elegans at different concentrations (20, 50, 100 μg/mL) to observe the effects of drugs on the motor ability and reproductive ability of nematodes. The changes of lipid content in Caenorhabditis elegans were determined by oil red O staining. The expression levels of skn-1, sbq-1, fasn-1 and fat-7 genes in Caenorhabditis elegans were detected by RT-PCR. Results: SQYSP alcoholic extracts improved Caenorhabditis elegans lifespan at both 50 μg/mL and 100 μg/mL concentrations (P < 0.05, P < 0.001), and SQYSP aqueous extracts improved nematode lifespan only at 100 μg/mL concentration (P < 0.01). In addition, both extracts could increase nematode body bending frequency (P < 0.01, P < 0.05). In the oil red O experiments, both alcoholic and aqueous extracts of SQYSP at high concentrations could improve the lipid accumulation in the nematode body (P < 0.001, P < 0.001), and the RT-PCR experiments showed that both extracts could increase the expression of skn-1 and decrease the expression of sbq-1, fasn-1, and fat-7 (P < 0.01). Conclusion: SQYSP has a lipid-lowering effect on Caenorhabditis elegans without damaging the normal physiological function of nematodes.
