%X 目的:灵芝具有抗阿尔茨海默病(AD)的潜力,但其作用机制尚不明确。方法:借用TCMSP检索筛选灵芝的化学成分及靶点,通过GeneCards数据库筛选出阿尔茨海默病的相关靶点,运用Cytoscape 3.6.0软件构建“药物–成分–疾病–靶点”网络图,运用String数据库构建靶点蛋白–蛋白相互作用(PPI)网络图。对药物–疾病的交集靶点进行基因本体(GO)及京都基因与基因组百科全书(KEGG)的富集分析,最后采用分子对接进行验证。结果:获得灵芝有效成分14个,AD疾病靶点4457个,灵芝与AD共同作用靶点22个,共得到GO分析104个,KEGG通路60条,主要涉及凋亡–多物种、胆碱能突触等。分子对接结果显示,核心成分与关键靶点具有良好的结合能力。结论:灵芝防治AD具有多成分、多靶点、多通路的特点。
Objective: Ganoderma lucidum has the potential to against Alzheimer’s Disease (AD), but its mechanism is still unclear. Methods: TCMSP was used to search and screen the chemical components and targets of Ganoderma lucidum, and the relevant targets of Alzheimer’s Disease were screened from the GeneCards database. The “drug component disease target” network diagram was constructed using the Cytoscape 3.6.0 software, and the target Protein-Protein Interaction (PPI) network diagram was constructed using the String database. The drug-disease intersection targets were enriched and analyzed by Gene Ontology (GO) and Kyoto Encyclopedia of Gene and Genome (KEGG), and finally verified by molecular docking. Results: 14 active components of Ganoderma lucidum, 4457 targets for AD disease, 22 targets for interaction between Ganoderma lucidum and AD, 104 GO analyses and 60 KEGG pathways were obtained, mainly involving apoptosis-multi-species, cholinergic synapses, etc. Molecular docking results show that the core components have a good binding ability with key targets. Conclusion: Ganoderma lucidum has the characteristics of multi-components, multi-targets and multi-pathways in the prevention and treatment of AD.
