The Isolation, Identification and Content Determination of Lupenone in the Flowers and Bracts of Musa basjoo, Musa paradisiacal, Wild Musa basjoo %A 余漂 %A 丁芹 %A 王祥培 %A 王远敏 %A 梁玉清 %A 吴红梅 %J Advances in Analytical Chemistry %P 1-12 %@ 2163-1565 %D 2022 %I Hans Publishing %R 10.12677/AAC.2022.121001 %X 目的:对芭蕉、香蕉及野蕉的花和苞片中羽扇豆酮进行分离与结构鉴定,并建立其含量测定方法。方法:利用硅胶柱色谱、质谱和核磁共振对芭蕉、香蕉及野蕉的花和苞片中的化合物进行分离纯化及结构鉴定;以高效液相色谱(HPLC)法对其含量进行测定。结果:从芭蕉、香蕉及野蕉的花和苞片中都能分离出羽扇豆酮;香蕉和野蕉的花中羽扇豆酮平均含量明显高于芭蕉花(P < 0.05),芭蕉苞片中羽扇豆酮平均含量明显高于香蕉和野蕉的苞片(P < 0.05);同植株芭蕉老花和嫩花中以老花中羽扇豆酮含量最高,芭蕉老苞片和嫩苞片中以老苞片的含量最高,三个品种中同植株花和苞片中羽扇豆酮含量均以苞片的较高。结论:本研究建立的含量测定方法简便、快捷、准确性高、重复性及专属性强,可用于不同品种香蕉、芭蕉及野蕉的花和苞片中羽扇豆酮的含量测定。提示芭蕉、香蕉及野蕉的花及苞片可作为抗糖尿病活性成分羽扇豆酮原药材来源。
Objective: To isolate and identify the lupenone in the flowers and bracts of Musa basjoo, Musa paradisiacal, Wild Musa basjoo, and establish a method to determine the contents. Method: The isolation and chemical structure identification of lupenone contained in the flowers and bracts of M. basjoo, M. paradisiacal, Wild M. basjoo were performed by silica gel column chromatography, mass spectrum and nuclear magnetic resonance; HPLC method was adapted to detect the contents of lupenone in the flowers and bracts of M. basjoo, M. paradisiacal, Wild M. basjoo. Result: The lupenone was isolated from the flowers and bracts of M. paradisiacal, M. basjoo, Wild M. basjoo. The average contents of lupenone in the flowers of M. paradisiacal and Wild M. basjoo were significantly higher than M. basjoo (P < 0.05), and the average contents of lupenone in the bracts of M. basjoo were significantly higher than M. paradisiacal and Wild M. basjoo (P < 0.05); the lupenone content in the old flowers was higher than that in the tender flowers of M. basjoo from same plant, and the lupenone content in the old bracts was higher than that in the tender bracts of M. basjoo from same plant. The lupenone content in bracts of M. paradisiacal, M. basjoo, Wild M. basjoo was higher than that in bracts of the same plant. Conclusion: The assay method established in this study is simple, rapid, accurate, repeatable and specific.
