%X 目的:胃腺癌作为常见癌症之一,虽然目前其诊疗方案日趋成熟,但其早期诊断及预后判断仍待完善。方法:本实验利用TCGA数据库筛选出胃腺癌潜在致病基因,并筛选相关hub基因,并做生存曲线。最后利用组织样本对上述结果进行验证。结果:利用TCGA数据库共筛选出29个潜在胃腺癌致病靶点,并筛选出10个关键基因。对关键基因进行数据分析,发现PTEN低表达及Villin高表达与胃癌患者生存率下降相关,最后利用免疫组化对现有标本进行验证。结论:PTEN低表达及Villin高表达可作为早期胃腺癌诊断的重要参考,其可能影响胃腺癌患者的预后。
Objective: Stomach adenocarcinoma is one of common cancer. Although its diagnosis and treatment plan is increasing, its early diagnosis and prognosis judgment remain treated. Methods: This experiment utilizes TCGA database to screen a potential pathogenic gene of gastric adenocarcinoma and screen the related HUB genes, and do survival curves. Finally, the above results are verified by tissue samples. Results: 29 potential gastric adenocarcinomas were screened using TCGA databases, and ten key genes were screened. Data analysis of key genes found that PTEN low expression and Villin high expression were related to the decrease in the survival rate of patients with gastric cancer. Finally, the existing specimens were verified using immunohistochemistry. Conclusion: PTEN low expression and Villin high expression can be an important reference for the diagnosis of early gastric adenocarcinoma, which may affect the prognosis of patients with gastric adenocarcinoma.
