The Technology System Preliminary Study of VIGS on Flax %A 陆旭鹏 %A 李世玉 %A 戴志刚 %A 袁红梅 %A 钱雨康 %A 粟建光 %A 孙健 %A 谢冬微 %J Botanical Research %P 499-506 %@ 2168-5673 %D 2021 %I Hans Publishing %R 10.12677/BR.2021.104063 %X 为研究烟草脆裂病毒(Tobacco rattle virus, TRV)介导的基因沉默(Virus-induced gene silencing, VIGS)技术是否适合在亚麻上应用,以PDS (Phytoene desaturase,八氢番茄红素脱氢酶)为标记基因,通过构建基因沉默载体,在亚麻品种大紫花中建立了TRV-VIGS体系。结果显示,pTRV2-PDS侵染植株真叶表现出明显的沿叶脉方向延伸的条状白化症状,且qRT-PCR分析发现与野生型及pTRV2空载体植物相比,pTRV2-PDS侵染的植株中PDS基因的表达均被有效抑制。证明,TRV介导的VIGS可以用于亚麻基因功能的验证研究。
In order to study whether tobacco rattle virus (TRV)-mediated virus induced gene silencing (VIGS) technology is suitable application for flax, a TRV-VIGS system is established by constructing a gene silencing vector with phytoene desaturase (PDS) as a marker gene. The results showed that qRT-PCR analysis showed that the expression of PDS gene in pTRV2-PDS infected plant is effectively inhibited compared with the control and empty plant, and the euphylla showed obvious albinism.
