%T LncRNA SNHG5对高糖缺氧环境下视网膜血管内皮细胞增殖及迁移的影响

%X 摘要 目的 探讨长链非编码RNA(LncRNA)小核仁RNA宿主基因5(SNHG5)对高糖缺氧环境下培养的人视网膜血管内皮细胞(HRCECs)凋亡和迁移能力的影响。 方法 将HRCECs分为5组,正常对照组(A组)培养于含5 mmol/L葡萄糖的DMEM培养基中,高糖组(B组)培养于含30 mmol/L葡萄糖的DMEM培养基中,高糖缺氧组(C组)培养于含体积分数0.01 O2以及30 mmol/L葡萄糖的DMEM培养基中,高糖缺氧过表达组(D组)将含有SNHG5目的基因的质粒转染进入HRCECs以后,培养于含有体积分数0.01 O2以及30 mmol/L葡萄糖的DMEM培养基中,高糖缺氧空载组(E组)将含有与D组等量的阴性对照质粒转染进入HRCECs后,与D组进行相同条件培养。采用实时荧光定量PCR(RT-qPCR)法检测各组细胞中SNHG5的表达量;采用Annexin V-FITC/PI双染流式细胞法检测各组细胞的凋亡百分比;采用细胞划痕法和Image J图像分析软件检测并计算各组细胞相对迁移面积。 结果 与A组比较,B组HRCECs中SNHG5基因的表达量降低(P<0.05),凋亡百分比增高(P<0.05),第36小时细胞的相对迁移面积增大(P<0.05);与B组比较,C组HRCECs中SNHG5的表达量降低(P<0.05),凋亡百分比增高(P<0.05),第36小时细胞相对迁移面积增高(P<0.05);与C组比较,D组HRCECs中SNHG5的表达量增高(P<0.05),凋亡百分比降低(P<0.05),第36小时细胞相对迁移面积降低(P<0.05);与E组比较,D组HRCECs中SNHG5的表达量增高(P<0.05),凋亡百分比降低(P<0.05),第36小时细胞相对迁移面积降低(P<0.05),且时间、组别及其交互作用均对HRCECs相对迁移面积有明显影响(F组别=252.93,F时间=791.36,F时间*组别=169.89,P<0.05)。 结论 高糖缺氧环境下培养可下调HRCECs中SNHG5的表达,过表达的SNHG5对HRCECs在高糖缺氧环境下引起的凋亡和迁移起明显的抑制作用。
Abstract:Objective To investigate the effect of the long non-coding RNA (LncRNA) small nucleolar RNA host gene 5 (SNHG5) on the apoptosis and migration of human retinal capillary endothelial cells (HRCECs) cultured in hyperglycemic and hypoxic conditions. Methods HRCECs were divided into normal control group (group A, cultured in DMEM medium containing 5 mmol/L glucose), hyperglycemic group (group B, cultured in DMEM medium containing 30 mmol/L glucose), hyperglycemic-hypoxic group (group C, cultured in DMEM medium containing 30 mmol/L glucose and O2 at a volume fraction of 0.01), hyperglycemic-hypoxic overexpression group (group D, the plasmid containing the SNHG5 gene was transfected into HRCECs, which were then cultured in DMEM medium containing 30 mmol/L glucose and O2 at a volume fraction of 0.01), and hyperglycemic-hypoxic empty-vector group (group E, the negative control plasmid with an equal amount as group D was transfected into HRCECs, which were then cultured in the same conditions as Group D). RT-qPCR was used to measure the expression of SNHG5 in each group of cells; Annexin V-FITC/PI staining and flow cytometry were used to measure the percentage of apoptotic cells; wound healing assay and Image J image analysis software were used to measure and calculate the relative migration area of cells in each group. Results Compared with group A, group B had a significant reduction in the expression of SNHG5 in HRCECs (P<0.05), a significant increase in the percentage of apoptotic cells (P<0.05), and a significant increase in the relative migration area of cells at 36 hours (P<0.05).
