The loss of function is impelled either by de-regulation of the E3 ubiquitin ligase, MDM2 protein (2) or by the TP53 gene mutations of missense type (3). Mutations in TP53 gene occur at a very high frequency and range from 10% to 97% depending on the cancer type (4).
