%T 烟酰胺磷酸核糖基转移酶在结肠癌中的表达、功能及生物信息学分析

%X 摘要 目的 探讨NAMPT在结肠癌组织中的表达并利用生物信息学分析其理化性质、蛋白结构、互作蛋白、细胞定位及对其功能进行预测分析。方法 利用TCGA数据库及即时聚合酶链锁反应（qPCR）技术验证NAMPT在结肠癌组织中的表达；利用siRNA敲除SW480细胞中NAMPT表达后测定细胞增殖能力；利用生物信息学ProtParam、Jpred4.0、Genecards、String等软件对NAMPT理化性质及功能进行预测分析。结果 在结肠癌组织中，NAMPT mRNA水平表达明显高于相应的癌旁组织；沉默SW480细胞中NAMPT后细胞增殖能力减弱；生物信息学提示NAMPT由491个氨基酸组成，为不稳定酸性亲水性蛋白质；二级结构预测发现其有14个α螺旋，17个β折叠；三级结构全球性模型质量估测（GMQE）评分0.97；预测结果显示NAMPT并无功能结构域，其仅有一个保守结构域。蛋白定位发现其在细胞外、细胞核以及胞质中均有大量表达，其参与的生物学活动主要包括参与NAD+生物合成、昼夜节律、应激、代谢反应及衰老等。结论 在结肠癌患者组织中NAMPT高表达，可参与多种生物学过程，并可通过不同途径调节肿瘤生物学行为
