金纳米棒由于其可变长径比,大比表面积和优良的光学特性已被广泛应用于生物分析领域。我们知道纳米金粒子是传统免疫层析分析技术中最为常用的一种探针,而金纳米棒在免疫层析分析技术中的应用报道鲜见报道。本文利用金纳米棒修饰单克隆检测抗体,构建新型信号探针,并成功应用于棉线为载体的免疫层析分析装置中,实现了肺癌标志物–人铁蛋白的快速,灵敏和定量检测。最优实验条件下线性范围为5~5000 ng/mL,其他蛋白质干扰很小。
Gold nanorods have been widely used in biological analysis due to their variable aspect ratio, large specific surface area and excellent optical properties. As we known, gold nanoparticle is the most commonly used probe in traditional immunochromatographic assay technology, while the application of gold nanorods in immunochromatographic analysis has rarely been reported. In this paper, gold nanorods modified with monoclonal antibody were used to construct a novel reporter probe, and applied in cotton thread immunochromatographic assay device successfully. The method has realized rapid, sensitive and quantitative detection of human ferritin (a lung cancer biomarker). Under optimal conditions, the linear range is 5 - 5000 ng/mL, and other proteins have little interference.
