%0 Journal Article %T 基于PCR-DGGE技术分析牧区酸马奶中乳酸菌多样性<br>Diversity of Lactic Acid Bacteria in Koumiss from Xinjiang and Inner Mongolia by PCR - DGGE %A 马俊英 %A 葛武鹏 %A 姜 %A 琪 %A 房若愚 %A 梁秀珍 %A 吴小勇 %A 耿 %A 炜 %J 西北农业学报 %D 2017 %R 10.7606/j.issn.1004-1389.2017.06.021 %X 采用聚合酶链式反应和变性梯度凝胶电泳(PCR-DGGE)技术,分析采自新疆和内蒙古6个牧区的混合酸马奶样品中乳酸菌的总DNA,利用PCR扩增其16S rRNA的V3区段,建立由标准菌株组成的参考梯度,对其进行PCR-DGGE,藉此鉴定菌株,对不能鉴定的菌株切胶回收进行测序鉴定,并分析比较优势菌株。结果表明:共鉴定出10种乳酸菌株,包括植物乳杆菌、嗜酸乳杆菌、马乳酒乳杆菌、发酵乳杆菌、詹氏乳杆菌、短乳杆菌、瑞士乳杆菌、乳酸乳球菌、嗜热链球菌及干酪乳杆菌;且嗜热链球菌为所有样品中的共有菌株;马乳酒乳杆菌、短乳杆菌及瑞士乳杆菌是新疆酸马奶中的优势菌株,而内蒙古样品中的优势菌株不尽相同。<br>After total DNA was extracted from koumiss samples from Xinjiang and Inner Mongolia,the V3 region of 16S rRNA gene was amplified by PCR,and a reference ladder derived from amplification products of known lactic acid bacteria was constructed at the same time,then the PCR amplified products were subject to identify the strain by using polymerase chain reaction and denaturing gradient gel electrophoresis(PCR-DGGE),the bands which could not be identified by reference ladder were excised from the gel and sequenced identification,so as to compare and analyze the dominant species.The results showed that 10 dominant species of lactic acid bacteria were identified from the samples.It included Lactobacillus plantarum,Lactobacillus acidophilus,Lactobacillus kefiranofaciens,Lactobacillus fermentum,Lactobacillus jensenii,Lactobacillus brevis,Lactobacillus helveticus,Lactococcus lactis,Streptococcus thermophilus and Lactobacillus casei; moreover,Streptococcus thermophiles appeared in all samples,the dominant species of samples in Xinjiang were Lactobacillus kefiranofaciens,Lactobacillus brevis and Lactobacillus helveticus while the dominant species were much different from the samples of Inner Mongolia. %K PCR-DGGE 乳酸菌 酸马奶 优势菌株< %K br> %K Polymerase chain reaction-denaturing gradient gel electrophoresis(PCR-DGGE) Lactic acid bacteria Koumiss Dominant species %U http://xbnyxb.alljournals.cn/ch/reader/view_abstract.aspx?file_no=20170621&flag=1
