%0 Journal Article %T 牦牛不同发育阶段睾丸中KDM2A的表达<br>Expression Analysis of KDM2A in Yak Testicle at Different Growth Phases %A 王 %A 艳 %A 熊显荣 %A 韩 %A 杰 %A 王 %A 斌 %A 杨显英 %A 阿果约达 %A 李 %A 键 %J 西北农业学报 %D 2018 %R 10.7606/j.issn.1004-1389.2018.09.003 %X 旨在研究 KDM2A在牦牛睾丸发育和精子发生中的作用,为提高牦牛繁殖性能提供理论依据。提取牦牛胎儿时期(5~6月龄)、幼年时期(1~2岁)、性成熟时期(3~4岁)睾丸的总RNA,通过实时荧光定量PCR和免疫组化检测 KDM2A在牦牛睾丸中的表达情况。结果显示: 3个时期牦牛睾丸中均有 KDM2A基因表达,且表达量随年龄增长呈递增趋势,性成熟时期睾丸中 KDM2A mRNA与蛋白质的表达量均显著高于胎儿时期和幼年时期;胎儿时期仅在精原干细胞及支持细胞中表达,幼年和性成熟时期睾丸中精原细胞、支持细胞、初级精母细胞、圆形精子及精子形成期均呈阳性。表明 KDM2A在牦牛不同发育阶段的睾丸中表达,在牦牛睾丸发育及精子生成中发挥重要作用。<br>The aim of this study was to reveal the role of KDM2A in testicle development and spermatogenesis of yak, and provide a theoretical basis for improving the breeding performance of yak. The total RNA was extracted from three stages of yak’s testicle, including fetus(5-6 month-old), juvenile period(1-2 year-old) and sexual maturity(3-4 year-old). The real time fluorescence quantitative PCR(RT-qPCR) and immunohistochemistry was used to detect the expression of KDM2A in yak testicle. The results showed that expression of the KDM2A gene has been observed in testicular tissue during three periods, and shown an upward trend with age. The expression levels of KDM2A mRNA and protein were significantly higher at maturity stage of yak testis than during fetus and juvenile period. The expression level of KDM2A protein was detected only in spermatogonial stem cells and sertoli cells of testicular tissues in fetal yak, and detected in spermatogonia cells, sertoli cells, primary spermatocytes, round sperm and spermiogenesis in testicular tissues of juvenile period and sexual maturity. The conclusion showed that KDM2A expressed at different growth period in yak testicular tissue, it suggested that KDM2A played an important role in testicle development and spermatogenesis, and made a foundation for the further study of the function of KDM2A gene in yak. %K 牦牛 KDM2A 睾丸 生精细胞< %K br> %K Yak KDM2A Testicle Spermatogenic cell %U http://xbnyxb.alljournals.cn/ch/reader/view_abstract.aspx?file_no=20180903&flag=1
