%0 Journal Article %T 马铃薯渣发酵饲料4种水解酶活性研究<br>Study on the Activity of Four Kinds of Hydrolases in Potato Slag Fermented Feed %A 任雅萍 %A 郭 %A 俏 %A 来航线 %A 陈姣姣 %A 薛泉宏 %J 西北农业学报 %D 2016 %R 10.7606/j.issn.1004-1389.2016.05.015 %X 采用固态发酵工艺与酶活性测定法研究发酵菌种、原料组成及原料灭菌处理对马铃薯渣发酵饲料中与动物饲料消化率密切相关的4种重要水解酶活性的影响。结果表明:向马铃薯渣中接入酵母菌(Yeast)、黑曲霉A8(Aspergillus niger A8)及米曲霉(Aspergillus oryzae)发酵剂能大幅度提高发酵产物蛋白酶、纤维素酶、果胶酶及植酸酶活性。加入尿素及油渣,进行原料灭菌处理也有类似效果:①发酵产物蛋白酶活性为184.20~564.90 U,发酵增率为191.7%~794.5%,接菌增率为68.9%~206.7%,灭菌增率为9.4%~ 45.2% ;②纤维素酶活性为3 455.56~5 978.15 U,发酵增率为4.7%~81.1%,接菌增率为36.7%~ 73.0% ,灭菌增率为1.7%~61.0%;③果胶酶活性为248.76~613.74 U,发酵增率为19.5%~194.8%,接菌增率为10.2%~146.7%,灭菌增率为7.5%~71.1%;④植酸酶活性为23.63~107.08 U,发酵增率为 67.7% ~ 949.6% ,接菌增率为109.4%~526.0%。除酵母菌接种的植酸酶外,其余4种水解酶接菌与不接菌的差异均达到显著水平(P<0.05)。单接米曲霉处理发酵产物的蛋白酶活性明显高于其他处理;单接黑曲霉及黑曲霉与酵母菌混接处理发酵产物的植酸酶活性均高于其他处理。<br>The study was conducted to investigate the effect of microbial inoculums, auxiliary materials and sterilization of material on the activity of four kinds of important hydrolases in solid-state fermentation product of potato slag with state fermentation and enzyme activity assay. The result showed that the activity of four kinds of hydrolases in the fermented potato slag increased significantly when added with microbial inoculums (Yeast, Aspergillus niger A8 and Aspergillus oryzae). Addition of oil foot and sterilization of material had a similar effect. In the fermentation product of sterilized material which contained oil foot especially, the activity of protease, cellulose, pectinase and phytase reached 184.20-564.90 U, 3 455.56-5 978.15 U, 248.76-613.74 U and 23.63-107.08 U, respectively. Compared with the blank control (unfermented sample), the increment rate of the activity of protease, cellulose, pectinase and phytase in the fermented potato slag were 191.7%- 794.5% , 4.7%-81.1%, 19.5%-194.8% and 129.6%-940.6%, respectively. Compared with the fermentation product without inoculation, the increment rate of the activity of protease, cellulose, pectinase and phytase in the fermented potato slag with microbial inoculation were 68.9%-206.7%, 36.7% -73.0%, 10.2%-146.7% and 109.4%-526.0%, respectively. The difference between the inoculation and non-inoculation treatments was significant except for the phytase activity in inoculation treatment with yeast(P<0.05). Compared with the fermentation product of unsterilized material, the increment rate of the activity of protease, cellulose, pectinase and phytase in the fermentation product of sterilized material were 9.4%-45.2%, 1.7%-61.0%, 7.5%-71.1% and 22.2%- 118.6% , respectively .
