%0 Journal Article %T α-烯醇化酶在肝癌组织中的表达及临床意义<br>Expression and clinical significance of α-enolase in hepatocellular carcinoma tissue %A 朱威威 %A 陈晓龙 %A 陈建安 %A 何玉婷 %A 余 %A 炎 %A 胡秋月 %A 孙冉冉 %A 任志刚 %A 李 %A 娟 %A 崔光莹 %A 张敏敏 %A 余祖江 %J 郑州大学学报(医学版) %D 2018 %R 10.13705/j.issn.1671-6825.2017.11.134 %X 目的:探究α-烯醇化酶(ENO1)在肝细胞癌(HCC)组织中的表达及临床意义。方法:利用TCGA和GEO公共数据库肝癌表达谱数据,分析HCC组织中ENO1 mRNA的表达; 依据ENO1 mRNA的表达水平将TCGA数据库中的肝癌样本分为ENO1 mRNA低表达组和高表达组,然后下载目标分析基因集进行富集分析; K-M法绘制生存曲线,比较两组总体生存期(OS)和无进展生存时间(PFS)。采用免疫组化染色方法检测93例HCC及87例癌旁正常组织中ENO1蛋白的表达; 将肝癌样本分为ENO1蛋白低表达组和高表达组,对两组进行生存分析。结果:HCC组织中ENO1 mRNA和蛋白表达水平均高于癌旁正常组织(P<0.05); TNMⅢ、Ⅳ期HCC组织中ENO1 mRNA和蛋白表达水平高于TNMⅠ、Ⅱ期组织(P<0.05),病理分级3、4级的HCC组织中ENO1 mRNA表达水平高于1、2级组织(P<0.05)。ENO1 mRNA低表达组中位OS和PFS均大于高表达组(P<0.05); ENO1蛋白低表达组中位OS大于高表达组(P<0.05)。与HCC预后不良相关的基因富集在ENO1 mRNA高表达组,与预后良好相关的基因富集在ENO1 mRNA低表达组。结论:ENO1与HCC的发生发展有关,有望成为HCC诊断及治疗的新靶点。<br>Aim: To explore the expression and clinical significance of α-enolase(ENO1)in hepatocellular carcinoma(HCC).Methods: The data of ENO1 mRNA expression in HCC tissue from TCGA and GEO common database were analyzed. According to the expression level of ENO1 mRNA, the HCC samples from the TCGA database were divided into ENO1 mRNA low expression group and high expression group; then the target analysis gene set was downloaded for GSEA analysis; K-M method was used to draw the survival curve, and the overall survival time(OS)and progression free survival time(PFS)of the two groups were compared. The expression of ENO1 protein in 93 HCC and 87 adjacent normal tissue was detected by immunohistochemical staining, the HCC samples were divided into ENO1 protein low expression group and high expression group, and the survival analysis about OS was carried out.Results: The expressions of ENO1 mRNA and protein in HCC tissue were higher than those in the normal tissue(P<0.05). The expression levels of ENO1 mRNA and protein in TNM Ⅲ and Ⅳ stage HCC tissue were higher than those in TNM Ⅰ and Ⅱ stage HCC tissue(P<0.05). The ENO1 mRNA expression in the pathological grade 3 and 4 of HCC tissue was higher than that in grade 1 and 2(P<0.05). The median OS and PFS of the ENO1 mRNA low expression group were higher than those of the high expression group(P<0.05), and the OS in the ENO1 protein low expression group was higher than that of the high expression group(P<0.05).
