%0 Journal Article %T 结肠炎小鼠外周血和结肠上皮组织中Gal??9的表达*<br>Expression of Gal??9 in peripheral blood and colonic epithelial tissue of mice with colitis %A 李治国 %A 江红艳 %A 李林静 %A 朱丽萍 %A 刘静 %A 冯百岁 %J 郑州大学学报(医学版) %D 2015 %X 摘要目的:探讨半乳糖凝集素??9(Gal??9)与炎症性结肠炎(IBP)的关系。方法:60只雄性Balb/c小鼠分为4组,每组15只。对照组:自由饮食。三硝基苯磺酸钠(TNBS)组:实验第1、8、15天分别用2.0、2.5和3.0 mg TNBS加体积分数50%的乙醇溶液100 μL灌肠,建立小鼠结肠炎模型。TNBS+鞭毛蛋白(CBir1)组:同上造模,并于实验第7、9、12、15天小鼠腹腔注射100 μg/L的CBir1 100 μL。TNBS+白细胞介素??17(IL??17)组:造模后,小鼠腹腔注射IL??17 500 ng,每周2次。第22天处死小鼠,ELISA方法检测外周血Gal??9含量,免疫组化法检测结肠上皮组织中Gal??9蛋白的表达。结果:各组小鼠血清和结肠上皮组织中Gal??9的表达差异有统计学意义(F/χ2=22.669和25.179,P均<0.001),TNBS组低于对照组,TNBS+CBir1组和TNBS+IL??17组低于TNBS组(P均<0.001)。结论:Gal??9可用来评估结肠炎的严重程度。<br>AbstractAim: To explore the pathogenic role of galectin??9(Gal??9) in colitis.Methods:Sixty male Balb/c mice were randomly divided into four groups.Control group:mice were given an unrestricted diet.TNBS group:mice were respectively instilled by 2.0, 2.5,and 3.0 mg TNBS dissolved in 100 μL 50% ethanol enema on the 1st,8th and 15th day of the experiment,by which TNBS??induced colitis models were established.TNBS+CBir1 groups:based on TNBS administration, CBir1 10 μg was dissolved in 100 μL saline, and each mouse was treated with an intraperitoneal injection of 100 μL of this solution during days 7,9,12, and 15.TNBS+IL??17 group: based on TNBS administration, each mouse was intraperitoneally injected with IL??17 twice a week. Then the mice were sacrificed on the 22nd day.The Gal??9 in peripheral blood and colonic epithelial tissue were detected by ELISA and immunohistochemistry respectively.
