%0 Journal Article %T 氯化锂联合hUC-MSCs移植对阿尔茨海默病小鼠的治疗作用<br>Effect of lithium chloride combined with hUC-MSCs transplantation on Alzheimer's disease mouse %A 孟 %A 楠 %A 马珊珊 %A 王欣欣 %A 姚 %A 宁 %A 邢 %A 衢 %A 宋及时 %A 黄团结 %A 关方霞 %J 郑州大学学报(医学版) %D 2016 %X 目的:研究氯化锂(LiCl)联合人脐带间充质干细胞(hUC-MSCs)移植对阿尔茨海默病小鼠(APP+转基因小鼠)的干预作用。方法:PCR鉴定为APP+转基因小鼠40只随机分为4组:APP+组、hUC-MSCs移植组、LiCl注射组、联合处理组(LiCl注射+hUC-MSCs移植)。处理1个月后,Morris水迷宫检测小鼠的认知水平,酶标仪法和分光光度法检测小鼠血清中超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量,qRT-PCR和Western blot法检测小鼠海马组织中Wnt/β-catenin通路相关因子的表达。结果:与APP+组比较,LiCl注射组APP+小鼠逃避潜伏期缩短,穿越平台次数增多,平台所在象限停留时间延长,MDA含量下降,小鼠海马组织中β-catenin、c-Myc mRNA和蛋白表达量增加,GSK-3β mRNA和蛋白表达量降低(P均<0.05); hUC-MSCs移植可使APP+小鼠血清中SOD活性升高(P均<0.05),其他指标的变化与LiCl注射组相同; LiCl注射联合hUC-MSCs移植对平台所在象限停留时间、MDA含量、c-Myc蛋白表达的影响具有协同效应(P<0.05)。结论:LiCl联合 hUC-MSCs移植对APP+转基因小鼠具有明显的治疗作用。<br>Aim: To research the effects of lithium chloride(LiCl)combined with human umbilical cord mesenchymal stem cells(hUC-MSCs)transplantation on Alzheimer's disease model mice(APP+ transgenic mice)and the potential mechanism.Methods: A total of 40 APP+ mice identified by PCR were allocated into 4 groups randomly:APP+ group, hUC-MSCs group, LiCl group and LiCl combined with hUC-MSCs group.After 1 month, all mice were to study the learning and memory ability by Morris water maze test, the activity of superoxide dismutase(SOD)and the concentration of malonaldehyde(MDA)in the serum were determined by microplate spectrophotometer and ultraviolet spectrophotometer, the mRNA and proteins associated with Wnt/β-catenin pathway were detected by qRT-PCR and Western blot.Results: Compared with APP+ group, the escaping latency was significantly shorter, the times of crossing platform was significantly increased and the time spent in the platform quadrant was significantly longer,MDA concentration was decreased,the expressions of β-catenin and c-Myc mRNA and proteins in the hippocampus tissue were increased, while GSK-3β mRNA and proteins expression was decreased in LiCl group; in hUC-MSCs group,the changing trend of the indexes mentioned above were similar with the LiCl group, and the activity of SOD in serum increased; hUC-MSCs transplantation and LiCl had synergistic effects on the time spent in the platform quadrant, MDA concentration and c-Myc protein expression(P<0.05).Conclusion: LiCl combined with hUC-MSCs transplantation has therapeutic effects on APP+ transgenic mice %K 氯化锂 %K Wnt/β-catenin通路 %K 干细胞移植 %K 阿尔茨海默病 %K 小鼠< %K br> %K lithium chloride %K Wnt/β-catenin pathway %K stem cell transplantation %K Alzheimer's disease %K mouse %U http://jms.zzu.edu.cn/oa/darticle.aspx?type=view&id=201602005
