%0 Journal Article %T 曲古霉素A对脊髓损伤小鼠的神经保护作用刘雯雯,程 田,马珊珊,黄团结,刘艳霞<br>Neuroprotective effects of trichostatin A on a mouse model of spinal cord injury %A 刘雯雯 %A 程 %A 田 %A 马珊珊 %A 黄团结 %A 刘艳霞 %A 程 %A 康 %A 李 %A 鹏 %A 许 %A 玲 %A 杨 %A 璐 %A 张 %A 涛 %A 关方霞 %J 郑州大学学报(医学版) %D 2018 %R 10.13705/j.issn.1671-6825.2017.06.144 %X 目的:探讨曲古霉素A(TSA)对脊髓损伤(SCI)小鼠的神经保护作用。方法:采用改良Allen's击打法制备SCI小鼠模型,随机分为TSA组和对照组(Vehicle 组,Veh组)(n=18),分别给予1 mg/kg TSA或等量生理盐水。术后第1、3、7、14、21、28天,BMS评分检测小鼠后肢运动功能。术后第3天,采用PI染色检测损伤附近区域坏死细胞数量,HEt染色检测活性氧(ROS)的累积; 心脏取血,采用ELISA法检测炎症因子TNF-α和IL-10的表达。术后第28天,qRT-PCR检测神经营养因子BDNF和NT3的表达量,免疫荧光染色检测MBP和NeuN的表达,记录白质损伤面积和进行神经元计数。结果:与Veh组相比,TSA组小鼠后肢运动功能恢复较好(P<0.05),小鼠损伤部位坏死细胞数减少,ROS含量减少, IL-10表达量升高而TNF-α表达量降低,BDNF和NT3表达量升高,损伤部位白质损伤面积显著降低,同时神经元数量增多(P均<0.05)。结论:TSA对SCI小鼠有神经保护的作用。<br>Aim: To investigate the neuroprotective effect of trichostatin A(TSA)on spinal cord injury(SCI)mouse model.Methods: A total of 36 mice were produced SCI mouse model by using modified Allen's method, and were randomly divided into Vehicle group(Veh group)and TSA group, 18 in each group. On the 3rd day after SCI, the number of necrotic cells near the injured area was detected by PI staining. HEt staining was used to detect reactive oxygen species(ROS)production. ELISA was used to detect the expressions of TNF-α and IL-10. On the 1st, 3rd, 7th, 14th, 21st and 28th day after operation, BMS score was used to detect the hindlimb motor function. On the 28th day after SCI, qRT-PCR was used to detect the expression of neurotrophic factor BDNF and NT3, and the expressions of MBP and NeuN were detected by immunofluorescence staining.Results: Compared with Veh group, hind limb motor function in TSA group was significantly improved, the number of necrotic cells and the production of ROS reduced, the expression of TNF-α decreased while expression of IL-10, BDNF and NT3 increased, the white matter(labeled by MBP)damage area decreased, and the number of neurons increased in the injury area(P<0.05).Conclusion: TSA plays a neuroprotective effect on a mouse model of SCI %K 脊髓损伤 %K 曲古霉素A %K 神经保护< %K br> %K 1)Stem Cell Laboratory %K School of Life Sciences %K Zhengzhou University %K Zhengzhou 450001 2)Department of Orthopedics %K the First Affiliated Hospital %K Zhengzhou University %K Zhengzhou 450052 %U http://jms.zzu.edu.cn/oa/darticle.aspx?type=view&id=201802003
