%0 Journal Article %T 脱氧雪腐镰刀菌烯醇的高灵敏化学发光磁酶免疫法检测*<br>Determination of deoxynivalenol by a sensitive method of magnetic enzyme chemiluminescence immunoassay %A 丁丽华 %A 于 %A 斐 %A 张冠军 %A 熊亚敏 %A 刘利娥 %A 吴拥军 %A 玉崧成 %J 郑州大学学报(医学版) %D 2017 %R 10.13705/j.issn.1671-6825.2017.02.007 %X 目的:建立一种快速简便、灵敏度高的新型化学发光磁酶免疫法(MECLIA)检测脱氧雪腐镰刀菌烯醇(DON)。方法:制备Fe3O4@SiO2@Au金磁复合微粒及DON与卵清蛋白(OVA)的复合物(OVA-DON); 以Fe3O4@SiO2@Au为固相载体,优化条件,使其表面负载OVA-DON形成DON金磁免疫复合微粒; 以鲁米诺-过氧化氢-辣根过氧化物酶-苯酚类化合物作为增强化学发光体系,建立MECLIA对小麦中DON进行检测,并对所建立的方法进行方法学评价。结果:该方法标准曲线方程为Y=-521 951X+2 883 979(X为DON质量浓度的对数,Y为化学发光值,R2=0.998 6),线性范围为0.10~20.00 μg/L,检测限为31 ng/L; 批内RSD为2.2%~3.5%,批间RSD为4.9%~9.6%,标准样品回收率为95.0%~108.0%,小麦样品回收率为92.0%~106.0%。结论:该研究建立的MECLIA灵敏度高、精密度好、操作简单,可用于大批量小麦样品中DON的快速检测。<br>Aim: To propose a magnetic enzyme chemiluminescence immunoassay(MECLIA)for rapid and sensitive detection of deoxynivalenol(DON).Methods: Fe3O4@SiO2@Au particles and the conjugation of DON to ovalbumin(OVA-DON)were prepared.Then,the DON-OVA was coated on the surface of Fe3O4@SiO2@Au by using Fe3O4@SiO2@Au as immobile phase. After that, MECLIA based on the luminol-H2O2-HRP-phenols system for detection of DON was established. The accuracy and sensitivity of the established method were evaluated.Results: The standard curve equation was Y=-521 951X+2 883 979(X was the logarithm of the concentration of DON,Y was the illumination value of the standard concentration, R2=0.998 6), the linear range was 0.10~20.00 μg/L, the limit of detection was 31 ng/L, the relative standard deviations were less than 3.5% and 9.6% for intra- and inter-assay, respectively, and the recoveries ranged from 95.0% to 108.0%.
