%0 Journal Article %T 实验性自身免疫性重症肌无力大鼠多肌群SNX17基因的表达<br>Analysis of SNX17 gene expression in multi-muscle group of active immunized EAMG rats %A 张艳停 %A 吕 %A 杰 %A 赵 %A 雪 %A 张迎娜 %A 方 %A 华 %A 李倩如 %A 杜 %A 英 %A 张清勇 %A 杨俊红 %A 张运克 %A 高 %A 峰 %A 李 %A 昕 %J 郑州大学学报(医学版) %D 2018 %R 10.13705/j.issn.1671-6825.2018.03.128 %X 目的:探讨实验性自身免疫性重症肌无力(EAMG)大鼠多肌群分拣微管连接蛋白17(SNX17)的表达特点。方法:采用主动免疫法构建EAMG大鼠模型(EAMG组)并进行评估,以正常大鼠作对照(正常对照组),利用荧光定量PCR和Western blot检测2组大鼠胸锁乳突肌、膈肌、咬肌、心肌、肋间肌、前肢肌、腓肠肌和眼肌等8组肌群中SNX17、nAChR mRNA和蛋白含量变化。结果:成功构建了EAMG大鼠模型。与正常对照组相比,EAMG组大鼠心肌、肋间肌中SNX17 mRNA的表达升高(P<0.05),膈肌中SNX17 mRNA的表达降低(P<0.05); 胸锁乳突肌、腓肠肌和眼肌中nAChR mRNA的表达降低(P<0.05),心肌中nAChR mRNA的表达升高(P<0.05); 咬肌、心肌和腓肠肌中SNX17蛋白含量升高(P<0.05),胸锁乳突肌、膈肌中降低(P<0.05)。EAMG组腓肠肌中SNX17 mRNA与 nAChR mRNA的表达呈负相关(r=-0.773,P=0.035),而在心肌中呈正相关(r=0.947,P=0.012)。结论:SNX17可能是神经肌肉接头处的关键蛋白。<br>Aim:To investigate the expression characteristics of sorting nexin 17(SNX17)in experimental autoimmune myasthenia gravis(EAMG)rats.Methods:EAMG rat model was constructed and evaluated by active immunization.Western blot and qRT-PCR were used to compare the changes of SNX17, nAChR contents in sternocleidomastoid, diaphragm, masseter, myocardial, intercostal muscle, forelimb muscle, gastrocnemius and eye muscle between EAMG group and control group.Results:The EAMG rats were successfully constructed. The expression level of SNX17 mRNA in the myocardial, and intercostal muscle of EAMG group was significantly higher than that of control group(P<0.05), and was lower in the diaphragm(P<0.05). The expression level of nAChR mRNA in the sternocleidomastoid,gastrocnemius and eye muscle of EAMG group was significantly lower than that of the control group(P<0.05),and was significantly higher in the myocardial muscle(P<0.05).The expression level of SNX17 protein in the masseter, myocardial, gastrocnemius of EAMG group was significantly higher than that of control group.The expression level of SNX17 protein in the diaphragm and sternocleidomastoid muscle was significantly lower than that of control group(P<0.05).The expression of SNX17 mRNA in the gastrocnemius muscle was negatively correlated with the expression of nAChR mRNA in the EAMG group(r=-0.773,P=0.035), and were positively correlated in the myocardial tissue(r=0.947,P=0.012).Conclusion:SNX17 may be a key protein in neuromuscular junctions %K 分拣微管连接蛋白17 %K 主动免疫 %K 实验性自身免疫性重症肌无力 %K 神经肌肉接头 %K 大鼠< %K br> %K sorting nexin 17 %K active immunization %K experimental autoimmune myasthenia gravis %K neuromuscular junction %K rat %U http://jms.zzu.edu.cn/oa/darticle.aspx?type=view&id=201806004
