%0 Journal Article %T TW37对食管癌TE-1细胞迁移的影响<br>Effects of TW37 on migration of esophageal cancer TE-1 cells %A 秦甜甜 %A 张雪燕 %A 李蕾蕾 %A 霍峻锋 %A 石晓丽 %A 王希雅 %A 叶小萍 %A 杨程宇 %A 王 %A 淙 %J 郑州大学学报(医学版) %D 2018 %R 10.13705/j.issn.1671-6825.2018.03.158 %X 目的:观察TW37对食管癌TE-1细胞迁移的影响。方法:用0.25、0.50、1.00、2.00、4.00、8.00、16.00、32.00和64.00 μmol/L的TW37处理TE-1细胞24、48、72 h,采用MTT法检测细胞增殖。倒置显微镜下观察2、4 μmol/L的TW37作用24 h对TE-1细胞形态的影响。划痕实验检测2、4 μmol/L的TW37作用24、48 h对TE-1细胞迁移的影响。Western blot实验测定2、4 μmol/L TW37作用24 h对TE-1细胞E-Cadherin、Vimentin以及N-Cadherin蛋白表达的影响。均设空白对照。结果:TW37抑制TE-1细胞增殖,呈时间和浓度依赖性。TW37作用后TE-1细胞由原来的梭形变成了卵圆形。TW37能够在较低浓度(2 μmol/L)抑制TE-1细胞迁移,4 μmol/L的TW37抑制迁移的作用更强(P<0.05)。TW37能降低TE-1细胞中Vimentin、N-Cadherin的表达,上调E-Cadherin的表达(P<0.05)。结论:TW37可能通过抑制EMT进程进而抑制TE-1细胞的迁移。<br>Aim:To observe the effects and possible mechanism of TW37 on the migration of esophageal cancer TE-1 cells.Methods:TE-1 cells were treated with 0.25,0.50,1.00,2.00,4.00,8.00,16.00,32.00,and 64.00 μmol/L TW37 for 24,48,and 72 h, respectively, and MTT assay was used to detect cell proliferation. TE-1 cells were treated by 2, 4 μmol/L TW37 for 24 h, the morphology of TE-1 cells was recorded by inverted microscope, and the expressions of E-Cadherin, Vimentin and N-Cadherin were measured by Western blot. TE-1 cells were treated by 2, 4 μmol/L TW37 for 24,48 h,respectively, and cell migration was detected by wound healing assay.Results:TW37 significantly inhibited the proliferation of TE-1 cells in a time-and concentration-dependent manner.TE-1 cells changed from the original shuttle into an oval-shape after being treated by TW37.TW37 could inhibit the migration of TE-1 cells at a lower concentration, which was stronger with higher concentration(P<0.05).
