%0 Journal Article %T iPSC-NSCs脑立体定向注射对脑出血大鼠脑血肿周围组织中IL-6、TGF-β1表达及神经 功能的影响<br>Effects of cerebral stereotaxic injection of iPSC-NSCs on expressions of IL-6, TGF-β1 around hematoma and neurological function in rats with intracerebral hemorrhage %A 秦 %A 洁 %A 马 %A 丹 %A 李 %A 珠 %A 孙石磊 %A 杨 %A 靖 %A 史长河 %A 宋 %A 波 %A 许予明 %J 郑州大学学报(医学版) %D 2018 %R 10.13705/j.issn.1671-6825.2017.07.107 %X 目的:探讨脑立体定向注射诱导多能干细胞来源的神经干细胞(iPSC-NSCs)对脑出血(ICH)模型大鼠神经功能及脑血肿周围组织中IL-6、TGF-β1表达水平的影响。方法:选取84只雄性SD大鼠采用大鼠脑立体定位仪制作ICH模型,然后分成3组:假手术组(仅定位穿刺,不注射胶原酶和PBS)、对照组(ICH模型+PBS)、实验组(ICH模型+PBS含iPSC-NSCs),每组28只。随后通过mNSS评分评价各组10只ICH模型大鼠在造模后第1、3、7、14、28天神经功能的动态变化。用ELISA方法检测各组ICH模型大鼠血肿周围组织中IL-6、TGF-β1的表达水平,每组每个时间点有6只大鼠。结果:不同处理方法的ICH 大鼠在不同时间点脑血肿周围组织中IL-6、TGF-β1的表达不同(P均<0.05)。实验组大鼠神经功能在第14、28天较对照组有明显改善。结论:iPSC-NSCs脑立体定向注射可调节ICH模型大鼠脑内主要促炎因子IL-6及抑炎因子TGF-β1的表达,并明显改善大鼠的神经功能。<br>Aim: To study the effects of cerebral stereotaxic injection of iPSC-NSCs on the expressions of IL-6 and TGF-β1 around hematoma and the neurological function in rats with intracerebral hemorrhage(ICH).Methods: A total of 84 ICH model rats were made using a rat brain stereotaxic instrument, and then randomly allocated into 3 groups, with 28 rats in each group: sham operation group(positioning the puncture without injecting collagenase or PBS), control group(ICH model+PBS), experimental group(ICH model+PBS containing iPSC-NSCs). Subsequently, the neurological functions of rats in each group were assessed respectively by mNSS after operation for 1, 3, 7, 14 and 28 d. The levels of IL-6 and TGF-β1 around the hematoma tissue were detected by ELISA after operation for 1, 3, 7 d,each group had 6 rats at each time point.Results: It was showed that the expressions of IL-6,TGF-β1 in the brain perihematoma tissue of ICH rats among 3 groups were statistically different at different time points(P<0.05). The neurological functions of rats in experimental group were significantly improved compared with those of the control group on the 14th and 28th day after ICH.Conclusion: Cerebral stereotaxic injection of iPSC-NSCs could improve the neurological functions of ICH model rats.
