%0 Journal Article %T 基于RNA-seq的杜氏盐藻全转录组测序与分析*<br>Transcriptome sequencing and analysis of Dunaliella salina based on RNA-seq %A 朱立强 %A 李庆华 %A 张彦婷 %A 朱相展 %A 关方霞 %A 薛乐勋 %J 郑州大学学报(医学版) %D 2016 %X 目的:利用RNA-seq技术对杜氏盐藻细胞的全部转录本进行测序、功能分析。方法:提取杜氏盐藻总RNA,反转录得cDNA,在Illumina平台上进行测序,经拼接、组装、聚类后获得全部unigene,并将所得unigene与数据库比对,对其进行功能注释和分类。结果与结论:共得到197 295个unigene,将获得的unigene与NR、SWISS-PROT、CDD、KEGG和TREMBI数据库进行比对,有89 800个unigene获得注释。其中95 767个unigene映射到GO的不同功能节点; 5 710个unigene获得COG注释,并分为22个功能分类; 9 497 个unigene获得了KEGG注释,映射到282条信号通路。所得杜氏盐藻的转录组信息为今后相关研究与应用提供了丰富的资源和线索。<br>Aim: To obtain the total transcripts of Dunaliella salina by RNA-seq and analyze their function.Methods: The total RNA of Dunaliella salina was extracted and reverse-transcribed to cDNA,which was sequenced on Illumina platform. The total unigenes were obtained after de novo assembly, and the unigenes were analyzed with functional annotation and classification after blasting with databases.Results and Conclusion: In the study, 197 295 unigenes were obtained, and 89 800 unigenes were annotated with gene descriptions, conserved protein domains, or GO terms against public databases NR, SWISS-PROT, CDD, KEGG and TREMBI. 95 767 of them were annotated with GO classification, 5 710 were grouped into 22 function categories compared against COG, and 9 497 annotated unigenes were assigned to 282 putative KEGG pathways.
