%0 Journal Article %T 转染miRNA-129-5p对MCF-7细胞化疗敏感性及ABCB1表达的影响<br>miRNA-129-5p transfection improves Taxol sensitivity on MCF-7 cells by regulating ABCB1 expression %A 石 %A 瑛 %A 王云凤 %A 路 %A 璐 %A 任静静 %A 马 %A 丹 %A 魏园玉 %J 郑州大学学报(医学版) %D 2018 %R 10.13705/j.issn.1671-6825.2017.06.077 %X 目的:探讨miRNA-129-5p转染是否可提高乳腺癌MCF-7细胞对紫杉醇的敏感性及其可能机制。方法:取对数生长期的MCF-7细胞,分别转染miRNA-NC和miRNA-129-5p,24 h后分别加入不同浓度的紫杉酸处理48 h,采用CCK-8法检测细胞增殖。根据细胞是否转染miRNA-129-5p和紫杉醇刺激培养将MCF-7细胞分为空白对照组、转染试剂组、转染miRNA阴性对照组(miRNA-NC组)、miRNA-NC+紫杉醇组、miRNA-129-5p组和miRNA-129-5p+紫杉醇组。利用流式细胞术检测细胞凋亡,采用RT-PCR和Western blot技术分别检测MCF-7细胞ABCB1、Bax和Bcl-2 mRNA及蛋白的表达。结果:与其他组比较,miRNA-129-5p转染细胞用紫杉醇处理后,MCF-7细胞增殖受抑,凋亡细胞增多,ABCB1和Bcl-2 mRNA及蛋白表达降低,而Bax mRNA和蛋白的表达升高(P均<0.05)。结论:miRNA-129-5p可能通过下调ABCB1的表达提高MCF-7细胞对紫杉醇的敏感性。<br>Aim: To explore the function of miRNA-129-5p transfection in improving Taxol sensitivity on MCF-7 cells.Methods: The MCF-7 cells were transfected with miRNA-NC and miRNA-129-5P,respectively, and then treated with different concentrations of Taxol for 48 h. The cell proliferation was detected by CCK-8.According to the cell transfection and culture protocol, MCF-7 cells were divided into six groups: blank control group, transfect reagent group, miRNA-negative control(NC)group, miRNA-NC+Taxol group, miRNA-129-5p group and miRNA-129-5p+Taxol group. The apoptosis was detected by flow cytometry. The expressions of ABCB1, Bax and Bcl-2 mRNA and protein were detected by RT-PCR and Western blot, respectively.Results: Cell proliferation was significantly inhibited, and cell apoptosis rate was higher in miRNA-129-5p+Taxol group(P<0.05). Simultaneously, compared with other groups, the mRNA and protein expressions of ABCB1 and Bcl-2 were significantly decreased(P<0.05), but mRNA and protein expressions of Bax were obviously increased in miRNA-129-5p+Taxol group(P<0.05).Conclusion: miRNA-129-5p might improve Taxol's drug sensitivity in breast cancer cells by inhibiting the expression of ABCB1 %K miRNA-129-5p %K ABCB1 %K 肿瘤耐药 %K 乳腺癌< %K br> %K miRNA-129-5p %K ABCB1 %K drug resistance %K breast cancer %U http://jms.zzu.edu.cn/oa/darticle.aspx?type=view&id=201802005
