%0 Journal Article %T miR??GAAB2??200a与金雀异黄酮对胃癌细胞生长及侵袭能力的影响<br>Synergistic effect of miR??GAAB2??200a and genistein on growth and invasion of gastric cancer cells %A 孙庆敏 %A 王 %A 进 %J 郑州大学学报(医学版) %D 2018 %R 10.13705/j.issn.1671-6825.2017.11.042 %X 目的:探讨miR??GAAB2??200a与金雀异黄酮对3种胃癌细胞生长及侵袭能力的影响及可能机制。方法:MGC803、BGC823、MKN45细胞分别分为4组:阴性对照组(转染阴性对照NC mimic)、金雀异黄酮组(加入50 μmol/L的金雀异黄酮)、miR??GAAB2??200a组(转染miR??GAAB2??200a mimic)、联合组(miR??GAAB2??200a mimic转染联合50 μmol/L的金雀异黄酮处理),MTT法观察处理24 h对细胞生长的影响,Transwell小室实验观察细胞侵袭能力的变化,RT??GAAB2??PCR及Western blot检测各组细胞EphA2 mRNA及蛋白的表达。结果:miR??GAAB2??200a与金雀异黄酮单用均可抑制部分胃癌细胞的生长、侵袭及EphA2蛋白的表达,两者交互作用仅出现在BGC823细胞中(P<0.05)。结论:miR??GAAB2??200a与金雀异黄酮可能通过调控EphA2抑制胃癌细胞生长及侵袭,且二者存在部分协同作用。<br>Aim: To investigate the effects and possible mechanisms of miR??GAAB2??200a and genistein on the growth and invasion of gastric cancer cells.Methods: The human gastric cancer cell lines MGC803, BGC823 and MKN45 were used as research objects. The cells were divided into 4 groups,negative control group(transfected with NC mimic),genistein group(treated by 50 μmol/L genistein),miR??GAAB2??200a group(transfected with miR??GAAB2??200a mimic),and combination group(transfected with miR??GAAB2??200a mimic and treated by 50 μmol/L genistein). The effects of miR??GAAB2??200a and genistein on the growth of gastric cancer cells were observed by MTT assay.Transwell chamber was used to verify the effect on cell invasion. RT-PCR and Western blot were used to observe the effects of miR??GAAB2??200a and genistein on EphA2 mRNA and protein.Results: Both miR??GAAB2??200a and genistein could inhibit the growth and invasion of part of the gastric cancer cells and inhibit the expression of EphA2 protein, and the synergistic effects of miR-200a and genistein were only found in BGC823 cells(P<0.05).Conclusion: miR??GAAB2??200a and genistein might inhibit the growth and invasion of gastric cancer cells through down-regulation of EphA2,and there are some synergistic effects %K miR GAAB2 200a %K 金雀异黄酮 %K EphA2 %K 胃癌< %K br> %K miR GAAB2 200a %K genistein %K EphA2 %K gastric cancer %U http://jms.zzu.edu.cn/oa/darticle.aspx?type=view&id=201804019
