%0 Journal Article %T Septin2在胃癌组织中的表达及对胃癌细胞增殖和凋亡的影响<br>Expression of Septin2 and its effect on cell proliferation and apoptosis in gastric cancer %A 李君君 %A 方 %A 磊 %A 高 %A 阳 %A 时 %A 帅 %A 何昊宇 %A 刘晓梅 %A 袁彩君 %J 郑州大学学报(医学版) %D 2018 %R 10.13705/j.issn.1671-6825.2017.11.012 %X 目的:研究GTP结合蛋白氯苄乙胺2(Septin2)在胃癌组织中的表达及对胃癌细胞增殖、凋亡的影响。方法:对基因表达综合数据库(GEO)中GSE54129数据集的基因芯片进行分析,比较胃正常组织和癌组织中Septin2 mRNA的表达差异; 利用Kaplan-Meier Plotter数据库进行生存分析,明确Septin2表达与胃癌预后的关系。收集胃正常和癌组织标本进行免疫组化染色,分析Septin2蛋白表达的差异。用Septin2基因表达质粒转染胃癌SCG-7901和MNK-45细胞,观察细胞增殖和凋亡情况。结果:胃癌组织中Septin2 mRNA和蛋白表达水平均高于正常组织(P<0.05),Septin2 mRNA表达水平与胃癌预后相关,Septin2 mRNA高表达者中位生存时间短于低表达者(P<0.001)。与空白对照组和阴性对照组相比,转染Septin2基因的SCG-7901和MNK-45细胞Septin2表达增加,细胞增殖能力增强,凋亡能力下降(P<0.001)。结论:Septin2基因过表达可促进胃癌细胞的增殖,抑制其凋亡; 与胃癌预后关系密切。<br>Aim: To investigate the Septin2 expression in gastric carcinoma tissue and its effect on proliferation and apoptosis of gastric cancer cells.Methods: The Septin2 mRNA expression in gastric cancer tissue and normal tissue from GSE54129 data set of GEO database were compared, and survival analysis was performed using the Kaplan-Meier Plotter database.The expressions of Septin2 protein in gastric normal tissue and cancer tissue were analyzed by immunohistochemical staining. The SCG-7901 and MNK-45 cell lines were transfected with the plasmid containing Septin2 gene, and then cell proliferation and apoptosis were analyzed.Results: The expression of Septin2 in gastric carcinoma tissue was significantly higher than that in normal tissue at mRNA level and protein level(P<0.001). Septin2 mRNA expression was associated with prognosis, and median survival time of patients with high Septin2 mRNA expression was shorter than that with low expression(P<0.001). The cells transfected Septin2 gene had higher proliferation abilities and lower apoptosis abilities(P<0.001).Conclusion: The overexpression of Septin2 gene can promote the proliferation and inhibit the apoptosis of gastric cancer cells.
