%0 Journal Article %T 表达US3基因腺病毒载体下调CTL和NK细胞对其转染肝细胞杀伤活性的影响<br>Attenuation on cytotoxic activities of T lymphocytes and NK cells against transgened hepatic cells by a recombinant adenovirus vector expressing US3 gene %A 王 %A 鹏 %A 李 %A 玲 %A 阚全程 %A 潘 %A 雪 %A 余祖江 %A 张振香 %A 冯 %A 婷 %J 西安交通大学学报(医学版) %D 2015 %R 10.7652/jdyxb201505016 %X 摘要:目的 研究表达US3基因重组腺病毒载体对其转染肝细胞介导的免疫逃逸活性。方法 首先构建表达US3基因腺病毒载体,扩增纯化后转染HL-7702肝细胞,利用细胞毒性T细胞(CTL)杀伤实验和自然杀伤细胞(NK)杀伤实验,检测表达US3基因重组腺病毒载体的免疫活性。结果 表达US3基因的重组腺病毒r-US3成功构建,r-US3能够明显降低CTL和NK细胞对转染肝细胞的杀伤活性。 结论 表达US3基因腺病毒载体在一定程度上能够介导转染肝细胞的免疫逃逸,降低机体免疫系统对转染肝细胞的排斥反应。<br>ABSTRACT: Objective To investigate the activity of immune evasion induced by a recombinant adenovirus vector expressing US3 gene in HL-7702 hepatic cells with transferred gene. Methods We first constructed an adenovirus vector expressing US3 gene and transferred it into HL7702 hepatic cells. Then immunological activities of US3 were determined by testing outivities of T lymphocytes (CTL) and natural killer (NK) cells. Results r-US3 and r-Track were successfully constructed. r-US3 could attenuate the activities of CTL and NK cells to HL-7720 cells with transferred gene.
