%0 Journal Article %T 豚鼠耳蜗血管纹TMEM16A随鼠增龄的表达变化<br>The expression of transmembrane protein 16A in guinea pig cochlea increases with years %A 宋 %A 佳 %A 王 %A 洋 %A 刘艳辉 %A 马克涛 %A 李 %A 丽 %A 司军强 %J 西安交通大学学报(医学版) %D 2017 %R 10.7652/jdyxb201706007 %X 摘要:目的 探讨跨膜蛋白16A(transmembrane protein 16A, TMEM16A)在不同鼠龄豚鼠耳蜗血管纹上的表达及其与年龄相关性听力下降之间的关系。方法 将健康豚鼠按照年龄分为:2周组、3月组、1年组、D-半乳糖(D-galactose, D-gal)衰老模型组;听性脑干反应(auditory brainstem response, ABR)检测不同鼠龄豚鼠的听力变化;免疫荧光技术和免疫印迹实验(Western blot)检测TMEM16A在不同鼠龄豚鼠耳蜗血管纹中的分布和表达改变。结果 ABR反应阈值随鼠龄增长呈逐渐增高趋势,D-gal组与其他3组之间的差异具有统计学意义(P<0.01)。TMEM16A在不同鼠龄豚鼠耳蜗血管纹中均有表达,且表达量从2周组到1年组随鼠龄的增加呈上升趋势(P均<0.05);D-gal组表达量减少,与3月组和1年组均有统计学意义(P<0.05)。结论 TMEM16A在豚鼠耳蜗血管纹表达的变化可能与年龄相关性听力下降相关。<br>ABSTRACT: Objective To investigate the expression of transmembrane protein 16A (TMEM16A) in the cochlea of guinea pigs and its relationship with the age-related hearing loss. Methods We used auditory brainstem response (ABR) to explore the changes of hearing in guinea pigs of different age (groups of 2w, 3m, 1y, and D-galactose). The distribution and expression of TMEM16A in the cochlea were detected by immunofluorescence and Western blot. Results ABR threshold was gradually increased, with significant difference between D-gal and the other three groups (P<0.01). TMEM16A was expressed in the cochlear striae vascularis at different ages, and the expression increased with age before 1y (P<0.05). However, its level was increased in D-gal group and significantly differed from that in 3m and 1y groups (P<0.05).
