%0 Journal Article %T 柠檬酚对人肝癌HepG2细胞增殖的抑制作用及其机制<br>Inhibitory effect and mechanism of citrusinol on proliferation of human hepatocellular carcinoma cells HepG2 %A 申恩华 %A 杜 %A 娟 %A 王立红 %A 张晶芬 %A 袁 %A 静 %A 付广双 %J 西安交通大学学报(医学版) %D 2017 %R 10.7652/jdyxb201704027 %X 摘要:目的 研究黄酮类化合物柠檬酚对人肝癌HepG2细胞增殖的抑制作用及其机制。方法 采用离体培养的HepG2细胞为研究对象,以MTT法检测细胞增殖抑制率;以HE染色和吖啶橙染色法观察细胞形态及分布;利用流式细胞仪对HepG2细胞的分裂周期进行检测;以荧光显色法检测蛋白表达。结果 柠檬酚能够剂量依赖性地抑制HepG2细胞的增殖,其抑制作用的IC50值为76.46μmol/L。柠檬酚能够使HepG2细胞发生皱缩,将细胞分裂阻滞在G2/M期,并抑制细胞骨架蛋白F-actin的表达。结论 柠檬酚具有抑制肝癌HepG2细胞分裂周期、抑制HepG2细胞骨架形成的作用,进而具有抑制肝癌HepG2细胞增殖的作用。<br>ABSTRACT: Objective To study the inhibitory effect and mechanism of citrusinol on proliferation of human hepatocellular carcinoma cells HepG2. Methods The inhibitory rate of HepG2 cells cultured ?? in vitro?? was measured by MTT assay. The morphology and distribution of the cells were observed by HE and acridine orange staining. The cell division cycle was detected by flow cytometry. The expression of F-actin protein was observed by fluorescent chromogenic method. Results Citrusinol could inhibit the growth of HepG2 cells, and the IC????50?? of the inhibitory rate was 76.46μmol/L. Citrusinol could make the HepG2 cells shrink, arrest the cell division cycle to G2/M, and inhibit the expression of F-actin.
