%0 Journal Article %T 铁碳纳米粒介导顺铂对肺癌细胞化疗敏感性的影响<br>Effects of carbon-coated iron nanoparticles carrying cisplatin on the chemotherapy sensitivity of lung cancer cells %A 党晓敏 %A 孙忠民 %A 杨 %A 岚 %A 尚 %A 东 %A 钟 %A 徽 %A 胡 %A 浩 %A 郭建新 %J 西安交通大学学报(医学版) %D 2017 %R 10.7652/jdyxb201704010 %X 摘要:目的 观察铁碳纳米粒搭载顺铂对肺癌细胞的抑制作用及对肺癌细胞Caspase 3和Survivin mRNA表达的影响。 方法 将肺癌细胞分为对照组、药物组、微粒组和药物微粒组,MTT法检测细胞生长活性,RT-PCR法检测肺癌细胞Caspase 3及Survivin mRNA的表达。结果 药物微粒组和药物组肺癌细胞的生长明显受抑制,尤其药物微粒组的抑制作用更为突出,且出现凋亡征象。药物组和药物微粒组肺癌细胞Caspase3 mRNA表达水平高于对照组,而Survivin mRNA表达减弱,并且药物微粒组的作用较药物组更为显著(P<0.05)。结论 铁碳纳米粒搭载顺铂能够显著增加肺癌细胞对顺铂的敏感性,增强药物的化疗效果。<br>ABSTRACT: Objective To investigate the inhibitory effect of carbon-coated iron nanoparticles carrying cisplatin on the growth of NCI-H446 lung cancer cells and expressions of Caspase 3 and Survivin mRNA. Methods NCI-H446 lung cancer cells were treated with iron-carbon nanoparticles and/or cisplatin. The cell viability was detected by MTT method, and the mRNA expressions of Caspase 3 and Survivin were measured with RT-PCR. Results Cisplatin could inhibit the growth of NCI-H446 lung cancer cells, and the inhibitory effect was stronger when it was combined with the iron-carbon nanoparticles. The cells had apoptosis. The mRNA expression of Caspase 3 of NCI-H446 lung cancer cells was remarkably enhanced after treatment with iron-carbon nanoparticles combined with cisplatin, while the mRNA expression of Survivin was notably weakened (P<0.05).
