%0 Journal Article %T NANOG在肝细胞癌中的表达及意义<br>Expression and clinical significance of NANOG in hepatocellular carcinoma %A 薛雨墨 %A 姚博文 %A 刘志奎 %A 徐 %A 蒙 %A 涂康生 %A 王 %A 军 %J 西安交通大学学报(医学版) %D 2016 %R 10.7652/jdyxb201605014 %X 摘要:目的 探讨NANOG在肝细胞癌(hepatocellular carcinoma, HCC)癌组织中的表达,并通过下调肝癌MHCC-97H细胞中NANOG的水平观察其对MHCC-97H细胞侵袭和迁移能力的影响。方法 收集89例HCC组织及对应的癌旁组织,应用免疫组化检测NANOG蛋白在肝癌及对应癌旁组织中的表达,并分析其表达与肝癌临床病理特征的关系。采用小干扰RNA(siRNA)下调肝癌MHCC-97H细胞中NANOG水平,Transwell实验检测MHCC-97H细胞的侵袭及迁移能力。结果 NANOG在肝癌组织中表达水平显著高于相应癌旁组织;肝癌组织中NANOG高表达与肿瘤大小、TNM分期、门静脉侵犯及Edmondson分级显著相关;NANOG高表达患者3年生存率明显低于低表达组;特异性siRNA下调NANOG水平后,明显抑制MHCC-97H细胞的侵袭和迁移。结论 NANOG在肝癌组织中的表达上调并与肝癌恶性临床病理特征有关,下调NANOG明显抑制肝癌细胞的侵袭和迁移能力。<br>ABSTRACT: Objective To detect the expression of NANOG in tumor tissue of human hepatocellular carcinoma and determine the effect of NANOG on cell invasion and migration in MHCC-97H cells by knockdown of NANOG. Methods We collected 85 pairs of HCC and para-carcinoma tissues and detected the expression of NANOG by immunohistochemical staining. Clinicopathological and prognostic significance of NANOG was evaluated in these patients. The expression of NANOG was knocked down by siRNA in MHCC-97H cells. Transwell assays were performed to test HCC cell invasion and migration in vitro. Results The expression of NANOG in HCC tissues was significantly higher than that in paired para-carcinoma tissues, and it was significantly associated with tumor size, TNM stage, portal vein infiltration and Edmondson degree. Patients in the higher NANOG group had a poorer 3-year survival than those in the lower group. When the expression of NANOG was downregulated by a specific siRNA, NANOG knockdown obviously inhibited cell invasion and migration in MHCC-97H cells. Conclusion NANOG is overexpressed in HCC tissue and is correlated with the malignant clinicopathologic features.
