%0 Journal Article %T 玉米南方锈病和普通锈病分子检测技术研究<br>Molecular detection of Puccinia polysora Underw. and P. sorghi Schw. %A 邢国珍 %A 魏馨 %A 李晶晶 %A 李闯 %A 刘娜 %A 蒋士君 %A 李春奇 %A 郑文明 %J 中国农业大学学报(自然科学中文版) %D 2017 %X 为建立快速鉴定玉米南方锈病菌和玉米普通锈病菌的分子检测方法,有助于玉米锈病的早期预警和有效防控,利用真菌ITS通用引物分别对采自海南、河南、吉林的玉米锈病菌样品进行了rDNA-ITS区克隆和测序分析(GenBank登录号为:HM452902,HQ154021~HQ154038),并根据其变异区段序列分别设计了病原菌检测特异性引物。结果表明:设计的检测引物具有较高的种特异性和检测灵敏度,在普通PCR体系中可检测到南方锈病菌和普通锈病菌的锈病菌DNA的最低浓度分别为100和1 pg/μL。本研究建立的检测体系对2种玉米锈病菌的快速分子鉴定及病害早期预警都具有重要应用价值。<br>The aim of this study was to develop molecular detection methods for rapid identification of corn rust pathogens,Puccinia polysora and P. sorghi,early warning and effective prevention of diseases caused.The DNA regions of rDNA-ITS of isolates,which were collected from Hainan,Henan and Jilin Provinces,were cloned and analyzed (GenBank accession number:HM452902,HQ154021-HQ154038) by using fungal IT Suniversal primers.Specific primers for the detection of P. polysora Underw. and P. sorghi Schw.were designed and tested accordingly.The primers showed high detection specificity and sensitivity for P. polysora Underw. and P. sorghi Schw.,respectively.The lowest pathogen DNA content can be detected in common PCR system was 100 and 1 pg/μL,respectively.Rapid detection technology developed in this study will benefit early prediction and effective control of corn rust. %K 玉米 玉米南方锈病 玉米普通锈病 分子检测< %K br> %K corn southern corn rust common corn rust molecular detection %U http://zgnydxxb.ijournals.cn/zgnydxxb/ch/reader/view_abstract.aspx?file_no=20170302&flag=1
