%0 Journal Article %T 大负荷运动及针刺干预对大鼠骨骼肌内质网应激钙信号的影响<br>Effects of Heavy Load Exercise and Acupuncture on Calcium Signal of Endoplasmic Reticulum Stressing in Skeletal Muscle of Rats %A 丁海丽 %A 孙竹昕 %A 王海牛 %A 任在方 %A 李俊平 %A 王瑞元 %J 北京体育大学学报 %D 2017 %R 10.19582/j.cnki.11-3785/g8.2017.11.009 %X 摘要:目的:探讨一次大负荷运动后大鼠骨骼肌内质网应激的钙信号机制,并观察针刺干预对其的影响。方法:8周龄雄性SD大鼠128只随机分为空白对照组(C)、单纯运动组(E)、单纯针刺组(A)、和运动针刺组 (EA)。其中E组和EA组进行一次大负荷运动,A组和EA组施以针刺干预。各组大鼠又按照运动后不同时间点分0 h、12 h、24 h、48 h和72 h组,在对应时相取比目鱼肌进行指标测试。使用TMEM法观测内质网钙离子浓度([Ca2+]ER)、 ELISA 法测定Ca2+-ATP酶(SERCA)含量;应用 Western Blot 法检测内质网应激蛋白GRP78、CRT表达。结果:1)一次大负荷运动后大鼠骨骼肌[Ca2+]ER急剧下降,SERCA含量亦明显下降;内质网应激蛋白 GRP78、CRT 表达明显上调,峰值出现在运动后即刻;2)针刺干预可使运动大鼠骨骼肌[Ca2+]ER和SERCA含量有所升高;GRP78和CRT蛋白表达相应下调。结论:一次大负荷运动可使骨骼肌内质网钙离子紊乱和相关酶功能障碍,进而诱导内质网应激的发生;运动后针刺可改善骨骼肌内质网功能受损,逐渐恢复[Ca2+]ER且有效调节内质网应激程度。<br>Abstract: Objective: To investigate the influence of heavy load exercise on calcium signal of endoplasmic reticulum stress in skeletal muscle of rats, and to observe the regulative effect of acupuncture. Methods: One hundred and twenty-eight 8-week old male SD rats were randomly divided into control group (C), simple exercise group (E), simple acupuncture group (A), and exercise + acupuncture group (EA). Group E and group EA carried out a heavy load downhill running, while group A and group EA were treated with acupuncture. The rats’ soleus muscle was obtained at 0h, 12h, 24h, 48h and 72h after exercise and tested. The concentration of Ca2+ ([Ca2+]ER) was determined by TMEM, the activity of Ca2+-ATPase (SERCA) was tested by ELISA, and the expression of GRP78 and CRT were detected by Western Blot. Results: 1) After heavy load exercise, [Ca2+]ER of skeletal muscle in rats decreased sharply and SERCA also decreased; GRP78 and CRT expressions increased, and their peak value appeared immediately after exercise; 2) acupuncture increased the activity of [Ca2+]ER and SERCA of skeletal muscle in exercised rats, and decreased GRP78 and CRT expressions. Conclusion: A heavy load exercise can cause Ca2+ disorders of endoplasmic reticulum and related enzyme dysfunction in skeletal muscle, and then induce endoplasmic reticulum stress; acupuncture can improve the impaired function of endoplasmic reticulum in skeletal muscle, gradually recover [Ca2+]ER and effectively regulate the level of endoplasmic reticulum stress %K exercise skeletal muscle endoplasmic reticulum stress calcium ion acupuncture %U http://xuebao.bsu.edu.cn/ch/reader/view_abstract.aspx?file_no=20171109&flag=1
