Wulongzhuyaozao %A 夏清清 %A 刘蓝筠 %A 孙海艳 %A 何桥 %A 李晓林 %A 梁国鲁 %A 朱恒星 %A 郭启高< %A br> %A XIA Qing-qing %A LIU Lan-jun %A SUN Hai-yan %A HE Qiao %A LI Xiao-lin %A LIANG Guo-lu %A ZHU Heng-xing %A GUO Qi-gao %J 西南大学学报(自然科学版) %D 2017 %R 10.13718/j.cnki.xdzk.2017.05.006 %X 以武隆猪腰枣为试材,以茎尖、带芽茎段、节间和叶片为外植体,开展了组织培养研究.结果表明,外植体以带芽茎段最好;适宜的初代培养基以1/2 MS+1 mg/L 6-BA+0.2 mg/L NAA,接种的带芽茎段萌芽快,其愈伤组织的不定芽再生率达53.3%;最佳不定芽继代培养基为MS+1 mg/L 6-BA+0.2 mg/L IBA,增殖系数达3.77;不定芽在1/2 MS+0.3 mg/L IBA和1/2 MS+0.3 mg/L IBA+10 g/L AC培养基上生根效果较好,生根率达90%~96.67%.<br>The shoot-tips, mature stem segments with buds, internodes and leaves were used as explants in a study of the tissue culture of jujube (<i>Zizyphus jujuba</i>) Mill. cv. Wulongzhuyaozao. The mature stem segments with buds were shown to be the most suitable explants. In the primary culture, 1/2MS+1 mg/L 6-BA+0.2 mg/L NAA was the best medium, in which the bud sprouted fast, and the rate of adventitious shoot regeneration from the induced calli was 53.3%. The best medium for proliferation was MS+1 mg/L 6-BA+0.2 mg/L IBA, and the bud multiplication coefficient was as high as 3.77. The adventitious shoots rooted readily on 1/2MS+0.3 mg/L IBA+10 g/L activated carbon, with a rooting percentage of 90%-96.67%. The identification of suitable conditions for tissue culture of Wulongzhuyaozao laid a good foundation for virus elimination, rejuvenation and variety improvement of this jujube cultivar %K 武隆猪腰枣 %K 组织培养 %K 快速繁殖< %K br> %K jujube (< %K i> %K Zizyphus jujuba< %K /i> %K ) Mill. cv.
