%0 Journal Article %T 半夏再生体系的建立<br>A Regeneration System of <i>Pinellia ternate</i> %A 吴能表 %A 曹瑞霞 %A 吴思婧 %A 杨怡< %A br> %A WU Neng-biao %A CAO Rui-xia %A WU Si-jing %A YANG Yi %J 西南大学学报(自然科学版) %D 2018 %R 10.13718/j.cnki.xdzk.2018.09.001 %X <sec>目的优化半夏再生条件,以获得优质半夏种苗. </sec> <sec>方法筛选出适宜的外植体后,使用3种激素2,4-D,KT,IAA利用正交设计法设置9个处理组对半夏愈伤组织进行诱导.选取长势良好,大小均一的半夏愈伤组织接于4种培养基中进行愈伤组织增殖.选取半夏叶柄诱导出的愈伤组织将其接于5种培养基中进行愈伤组织分化,观察不同培养基对半夏愈伤组织生根和出芽的影响,最后进行炼苗与移栽. </sec> <sec>结果在叶柄、叶片及块茎3种外植体中,叶柄是半夏愈伤组织诱导的最佳外植体;最有利于半夏愈伤组织诱导的培养基为MS+0.5 mg/L 2,4-D+1.5 mg/L KT+1.0 mg/L IAA;在暗培养条件下,将半夏愈伤组织继代于MS+2.0 mg/L 2,4-D+1.0 mg/L KT+1.0 mg/L IAA中生长最快;MS+0.2 mg/L 2,4-D+1.5 mg/L 6-BA培养基最有利于半夏愈伤组织的生根和出芽. </sec><br><sec>ObjectiveThis study was conducted to build a regeneration system of <i>Pinellia ternate</i> and optimize the regeneration conditions so as to obtain high-quality plantlets for cultivation. </sec> <sec>MethodsAfter screening out suitable explants of <i>P. ternate</i>, the experiment materials were divided into nine treatment groups according to the orthogonal design and treated with 2, 4-D, KT and IAA to induce calli. Uniform healthy calli of <i>P. ternate</i> were selected and inoculated to four multiplication media. Petiole-derived calliwere selected and inoculated to five differentiation media. Then, the effects of different media on the rooting and sprouting of <i>P. ternate</i> calli were observed. Finally, the regenerated plantlets were hardened and transplanted. </sec> <sec>ResultsOf the three explants studied (petiole, leaf and tuber), petiole was shown to be the best explant for callus induction. The best callus induction medium of <i>P. ternate</i> was MS+0.5 mg/L 2, 4-D+1.5 mg/L KT+1.0 mg/L IAA. When culture was made in the dark, the best callus induction medium of <i>P. ternate</i> was MS+2.0 mg/L 2, 4-D+1.0 mg/L KT+1.0 mg/L IAA.
