%0 Journal Article %T 基于木瓜蛋白酶过氧化物酶活性检测谷胱甘肽<br>Colorimetric Detection of Glutathione Based on Peroxidase-Like Activity of Papain %A 庞妍娇 %A 于媛媛 %A 郑鹄志 %A 隆异娟< %A br> %A PANG Yan-jiao %A YU Yuan-yuan %A ZHENG Hu-zhi %A LONG Yi-juan %J 西南大学学报(自然科学版) %D 2017 %R 10.13718/j.cnki.xdzk.2017.09.019 %X 木瓜蛋白酶是新发现的具有过氧化物模拟酶活性的蛋白酶,它能催化H<sub>2</sub>O<sub>2</sub>氧化3,3',5,5'-四甲基联苯胺(TMB),发生蓝色反应,并在652 nm处有吸收峰.当引入谷胱甘肽(GSH)后,由于GSH消耗H<sub>2</sub>O<sub>2</sub>,使其在652 nm处吸光度降低,且溶液颜色变浅.基于此,设计了一种方便简单、灵敏的比色分析法检测GSH,在生物样品检测中具有潜在的应用.<br>Papain, a newly discovered proteinase, possesses peroxidase enzyme activity, which can catalyze the oxidation of the peroxidase substrate 3, 3', 5, 5'-tetramethylbenzidine (TMB) in the presence of H<sub>2</sub>O<sub>2</sub> to the oxidized TMB to induce a blue colored reaction with a typical absorption peak centered at 652 nm. However, GSH can consume H<sub>2</sub>O<sub>2</sub>, which results in a fading of the blue color of the reaction system and a decrease of the absorbance. Accordingly, GSH can be detected by measuring the wastage of H<sub>2</sub>O<sub>2</sub>.
