%0 Journal Article %T 基于富硒条件下高温型香菇的液体培养基优化<br>Optimizing the Liquid Medium of High Temperature Type <i>Lentinus edodes</i> Under Se-Enriched Condition %A 乔艳明 %A 柳林 %A 陈文强< %A br> %A QIAO Yan-ming %A LIU Lin %A CHEN Wen-qiang %J 西南大学学报(自然科学版) %D 2018 %R 10.13718/j.cnki.xdzk.2018.03.005 %X 筛选适宜高温型香菇菌丝生长的Na<sub>2</sub>SeO<sub>3</sub>质量浓度, 优化其最佳的富硒液体发酵培养基, 拟为富硒香菇的相关产品开发提供参考依据.以高温型香菇Q12菌株为试材, 根据菌丝生长状况确定最佳的Na<sub>2</sub>SeO<sub>3</sub>质量浓度, 通过Plackett-Burman设计试验、最陡爬坡试验、Box-Behnken响应面分析试验及验证试验得到其在富硒条件下的最佳液体发酵培养基.结果表明:富硒条件下香菇Q12菌株液体发酵的最佳培养基组合:玉米粉39.74 g、红糖15.70 g、牛肉粉5.00 g、麦麸15.00 g、KH<sub>2</sub>PO<sub>4</sub> 0.80 g、MgSO<sub>4</sub>·7H<sub>2</sub>O 1.00 g、Na<sub>2</sub>SeO<sub>3</sub> 30.00 mg/L, 利用该培养基发酵得到的香菇菌丝生物量为57.56 g/L, 比优化前提高了1.30倍.数学模型的预测值与试验观察值相符, 相对误差约为5.95%, 响应面法优化富硒条件下高温型香菇的液体发酵培养基可行.<br>In order to provide reference for the development of shiitake mushroom-related products, the most suitable concentration of Na<sub>2</sub>SeO<sub>3</sub> for the mycelial growth of the high temperature type shiitake mushroom (<i>Lentinus edodes</i>) strain Q12 was selected and its Se-enriched liquid medium was optimized.Through the experiments of initial experimental design of Plackett-Burman, path of steepest ascent, Box-Behnken design and response surface analysis, the optimum liquid fermentation medium of 'Q12' under Se-enriched condition was shown to be corn meal 39.74 g + brown sugar 15.70 g + powdered beef 5.00 g + wheat bran 15.00 g + KH<sub>2</sub>PO<sub>4</sub> 0.80 g + MgSO<sub>4</sub>·7H<sub>2</sub>O 1.00 g + Na<sub>2</sub>SeO<sub>3</sub> 30.00 mg/L.On this optimized fermentation medium, the mycelial biomass of 'Q12' was as high as 57.56 g/L, which was 1.30 times higher than that on the un-optimized medium.The predicted values of a mathematical model were consistent with the experimental observations, the relative error being about 5.95%, and it was, therefore, concluded that the response surface method is feasible for the optimization of Se-enriched liquid medium of high temperature type <i>L.edodes</i> strains %K 香菇 %K 富硒 %K 高温型 %K 液体培养基 %K 响应面法< %K br> %K < %K i> %K Lentinus edodes< %K /i> %K Se-rich %K high temperature type %K liquid medium %K response surface method %U http://xbgjxt.swu.edu.cn/jsuns/html/jsuns/2018/3/201803005.htm
