%0 Journal Article %T TS表达与MMR状态联合检测在结直肠癌预后中的研究进展 %A 张西综述 %A 许青审校 %J 同济大学学报(医学版) %D 2015 %R 10.16118/j.1008-0392.2015.01.029 %X 胸苷酸合成酶(thymidylate synthetase, TS)是5-Fu(5-fluorouraci)发挥抗代谢作用治疗胃肠道恶性肿瘤的重要作用靶点,TS mRNA及蛋白表达与5-FU类药物的敏感性和抗药性密切相关。DNA错配修复MMR蛋白缺乏可以引起微卫星不稳定(microsatellite instability,MSI)。MSI亚型与5-FU类药物的疗效及结直肠癌患者预后相关。目前,将TS表达或MMR状态单独作为结直肠癌预后因子仍存在争议,因此联合检测TS表达与MMR状态 来判断结直肠癌患者预后的研究受到越来越多的关注。</br>Thymidylate synthase (TS) is an important target of 5-fluorouracil(5-FU)in anti-metabolic therapy of gastrointestinal malignancies. The expression of TS mRNA and TS protein are closely related to sensitivity of 5-FU-based chemotherapy. Microsatellite instability (MSI) can be caused by deficient DNA mismatch repair(MMR). MSI was reported to be associated with the efficacy of 5-Fu-based chemotherapy and prognosis of patients. It is questionable whether a single marker of TS or MMR might play a relevant prognostic and predictive role because colorectal cancer is a complex and heterogeneous disease.
