%0 Journal Article %T 敲降TMEM45A基因对卵巢癌细胞增殖、黏附及侵袭的影响 %A 门传娣 %A 任青 %A 郭静 %A 罗宁 %A 杨伟红 %A 程忠平 %J 同济大学学报(医学版) %D 2018 %R 10.16118/j.1008-0392.2018.01.010 %X 目的 观察跨膜蛋白45A(TMEM45A)基因对卵巢癌细胞增殖、侵袭能力的调节作用,并进一步探索其可能相关信号通路。方法 通过分析TCGA数据库卵巢癌组RNA测序数据和25对卵巢癌及其配对非癌组织样本的RT-PCR数据,比较卵巢癌组织和正常组织中TMEM45A的表达。在两种卵巢癌细胞系HO-8910和A2780中进行RNA干扰,抑制TMEM45A的表达;使用CCK-8、PI染色、细胞黏附和Transwell实验检测细胞增殖、细胞周期分布、黏附和侵袭能力;分析TGF-β信号传导途径、粘着斑途径与TMEM45A的高表达的相关性;采用RT-PCR和Western检测TGF-β1、TGF-β2、RhoA、ROCK2 mRNA和蛋白水平。结果 TMEM45A在卵巢癌中过表达(P<0.001);TMEM45A基因的沉默抑制了细胞增殖(P<0.05),增加了处于G1期的细胞群(P<0.05);敲降TMEM45A基因可以抑制细胞黏附和侵袭(P<0.05);抑制TMEM45A显著下调TGF-β1/2、RhoA和ROCK2的表达(P<0.05)。结论 TMEM45A可能是卵巢癌的致癌基因,敲降TMEM45A基因可能成为卵巢癌的治疗靶点。</br>Objective To investigate the effect of TMEME45A gene on the proliferation and invasion of ovarian cancer cells and related signaling pathway. Methods The expression of TMEM45A was compared between ovarian cancer and normal tissues based on the RNA-sequencing data of the ovarian cancer cohort from The Cancer Genome Atlas (TCGA) project. The expression of TMEM45A was also detected by RT-PCR in 25 pairs of ovarian cancer tissues and the matched non-cancerous tissues. The expression of TMEM45A in ovarian cancer HO-8910 and A2780 cells was down-regulated by RNA interference. Cell proliferation, cell cycle distribution, cell adhesion and invasion ability were detected by Cell Counting Kit-8 assay (CCK-8), propidium iodide (PI) staining, cell adhesion and Transwell assays, respectively. In addition, the mRNA and protein expression levels of transforming growth factor-β (TGF-β1 and TGF-β2), Ras homolog family member A (RhoA) and Rho-associated kinase 2 (ROCK2) were detected by RT-PCR and Western blotting, respectively. Results TMEM45A was over-expressed in ovarian cancer(P<0.001). Silence of TMEM45A suppressed cell proliferation(P<0.05)and increased cell population in the G1 phase significantly(P<0.05). Knock-down of TMEM45A inhibited cell adhesion and invasion(P<0.05), down-regulated the expression of TGF-β1/2, RhoA and ROCK2(P<0.05).
