%0 Journal Article %T 磷脂酶D1对胰腺癌细胞增殖的影响 %A 饶倩雯 %A 王菲 %A 宋美怡 %A 杨长青 %J 同济大学学报(医学版) %D 2018 %R 10.16118/j.1008-0392.2018.03.003 %X 目的 探究磷脂酶D1(PLD1)对于胰腺癌细胞增殖的影响。方法 通过CCK-8法、EdU荧光染色和流式细胞术检测PLD、PLD1及PLD2抑制剂对胰腺癌细胞系Capan-2细胞活力、增殖、周期的影响;采用同样方法检测抑制/过表达PLD1对Capan-2细胞活力、增殖、周期的影响。结果 PLD、PLD1抑制剂均抑制了Capan-2增殖(P=0.004、0.044),而PLD2抑制剂对Capan-2增殖无明显影响(P=0.945)。下调PLD1的表达使Capan-2细胞活力降低(P=0.000)、增殖能力减弱(P=0.002),阻碍了细胞周期进展(P=0.004、0.002);上调PLD1的表达使Capan-2细胞活力提高(P=0.004)、增殖能力增强(P=0.002),促进细胞周期进展(P=0.0004、0.035)。结论 PLD1能够促进胰腺癌细胞的增殖,在胰腺癌的发展中发挥着重要的作用。</br>Objective To explore the role of PLD1 in regulating proliferation of pancreatic cancer cells. Methods The effects of PLD, PLD1 and PLD2 inhibitors on the cell viability, proliferation and cell cycle of pancreatic cancer were detected by CCK-8, EdU fluorescent staining and flow cytometry, respectively. Furthermore, the effects of PLD1 knockdown or overexpression on the cell viability, proliferation and cell cycle of Capan-2 cells were detected. Results Both PLD and PLD1 inhibitors inhibited the proliferation of Capan-2 cells(P=0.004,0.044), while PLD2 inhibitors did not have significant effects on the proliferation of Capan-2 cells(P=0.945). Downregulation of PLD1 decreased the cell viability(P=0.000), cell proliferation(P=0.002) and suppressed cell cycle progression of Capan-2 cell(P=0.004,0.002), while upregulation of PLD1 enhanced the cell viability(P=0.004), cell proliferation(P=0.002) and promoted cell cycle progression of Capan-2 cell(P=0.0004,0.035).
