%0 Journal Article %T 外源水杨酸处理对活血丹中次生代谢产物积累的影响 %A 刘丽 %A 李姿 %A 马雪莲 %A 郭巧生 %A 窦道龙 %J 南京农业大学学报 %D 2016 %R 10.7685/jnau.201508041 %X [目的] 研究外源水杨酸(SA)处理对活血丹中总黄酮、齐墩果酸和熊果酸等次生代谢产物含量的影响,进一步优化其施用方法,以完善活血丹规范化种植技术,保障药材品质。[方法] 以野生活血丹扦插苗为试验材料,研究SA处理前后活血丹中总黄酮、齐墩果酸和熊果酸含量的变化,从而探讨SA处理与活血丹中总黄酮、齐墩果酸和熊果酸含量变化是否具有相关性,并在此基础上,比较不同外源SA施用浓度(0、7、14、21和28 mg?L-1)、不同处理时间(4、7、10、13、16和19 d)及施用次数(0、1、2、3和4次)下,活血丹生长及其中总黄酮、齐墩果酸和熊果酸含量的变化。总黄酮采用超声提取法提取,分光光度法测定含量,齐墩果酸与熊果酸采用超声法提取,HPLC法测定含量。[结果] 外源SA喷施处理后,活血丹中总黄酮、齐墩果酸和熊果酸的含量随SA质量浓度升高呈现先升后降的趋势,当外源SA质量浓度为14 mg?L-1时,三者的含量达到最大值。当外源SA浓度相同时,活血丹中总黄酮、齐墩果酸和熊果酸的含量总体上呈现先升后降的趋势,但均较同期对照组显著增加,并多在处理13 d达峰值。相同浓度下喷施1~4次SA溶液,均可使活血丹中总黄酮、齐墩果酸和熊果酸含量增加。[结论] 外源水杨酸处理可显著提高活血丹中总黄酮、齐墩果酸和熊果酸含量。最优的处理条件为:质量浓度为14 mg?L-1的水杨酸溶液每4 d喷施1次,共喷施4次,最后一次喷施在采收前12~14 d。</br>[Objectives] Effects of salicylic acid(SA)application on the contents of total flavonoids, oleanic acid, and ursolic acid in Glechoma longituba were evaluated to optimize cultivation technique and guarantee medicinal quality.[Methods] Wild cutting seedlings of G. longituba were treated with SA at different concentrations(0, 7, 14, 21, 28 mg?L-1), days after treatment(4, 7, 10, 13, 16 and 19 d)and different treatment times(0, 1, 2, 3, 4). Untrasonic extraction was employed to extract total flavonoids, oleanic acid, and ursolic acid. The content of total flavonoids was determined by the spectrophotometry, and contents of the oleanic acid and ursolic acid were measured by HPLC.[Results] Contents of total flavonoids, oleanic acid, and ursolic acid were initially increased and then decreased with the increasing exogenous SA concentrations, and peaked at the concentration of 14 mg?L-1. Under the same SA concentration condition, contents of total flavonoids, oleanic acid, and ursolic acid initially increased and then decreased with the extension of days after application, and the highest contents were all obtained in 13 day after treatment. SA treated for 1-4 times led to increase the contents of total flavonoids, oleanic acid, and ursolic acid. The treatment especially the content of total flavonoids, and treatment with SA for one time couldn’t lead to a significant improvement of their contents.[Conclusions] Exogenous SA treatment was demonstrated to increase the contents of total flavonoids, oleanic acid, and ursolic acid in G.longituba.
