%0 Journal Article %T 菊花不同时期各组织器官石蜡切片制作条件的优化 %A 刘涛 %A 任莉萍 %A 曹沛沛 %A 陈发棣 %A 房伟民 %A 陈素梅 %A 管志勇 %A 腾年军 %A 张飞 %A 赵爽 %A 王海滨 %A 宋爱萍 %A 蒋甲福 %J 南京农业大学学报 %D 2016 %R 10.7685/jnau.201601030 %X [目的] 根据菊花不同时期各组织器官特性,设计试验对石蜡切片制作条件进行优化,旨在筛选针对特定组织的节省时间和成本且效果最佳的石蜡切片制作条件。[方法] 取不同时期菊花根、茎、叶、花等样品,采用常规石蜡切片制作方法并结合试验过程中总结的固绿快速染色法,设计不同的渗蜡时间、切片厚度及染色方法,对各组织器官进行石蜡切片制作,经显微拍照后筛选出各组织最适石蜡切片的制作条件。[结果] 经筛选发现,在渗蜡时间方面,渗蜡充分幼根需5 d,老根6 d,幼茎6 d,老茎7 d,叶片4 d,花芽5 d,舌状花4 d。在切片厚度方面,最适切片厚度幼根为16 μm,老根20 μm,幼茎12 μm,老茎12 μm,叶片25 μm,花芽10 μm,舌状花16 μm。在染色方法的选择方面,幼根经番红-固绿双重染色后效果最佳,老根则首选番红-固绿双重染色法或番红单染法;幼茎和老茎同样首选番红-固绿双重染色法或番红单染法;叶片、花芽及舌状花以操作最为简单的固绿快速染色法为首选。[结论] 试验针对菊花不同时期各组织器官所筛选的最优石蜡切片制作条件,能够最大限度地节省时间和成本,并能达到试验观察所需的最优效果。</br>[Objectives] According to the characteristic of different tissues in different period,the experiment is designed to optimize the process of paraffin section making. In other words,it is designed to select specific paraffin section making method for specific tissues that can save time and cost. [Methods] For the paraffin section making of chrysanthemum different tissues,experiment was conducted with chrysanthemum different tissues,such as roots,stems,leaves,flowers that come from field planting early days,flower differentiation and blooming stage as material for paraffin sectioning. Then use different permeability wax time,section thickness and staining method were used to make paraffin sectioning. [Results] After repeated screening,we found in the aspect of permeability wax time,5 d is enough for young root,old root needs 6 d,young stem needs 6 d,old stem needs 7 d,leaf needs 4 d,flower bud needs 5 d,ray flower needs 4 d;in the aspect of section thickness,16 μm is best for young root,old root needs 20 μm,young stem needs 12 μm,old stem needs 12 μm,leaf needs 25 μm,flower bud needs 10 μm,ray flower needs 16 μm;in the aspect of staining method selection,sarranine and fast green staining method or sarranine taining method is the best method for young root;sarranine and fast green staining method or sarranine staining method are best for old root,young stem and old root;fast green quickly staining method is the best for leaf,flower bud and ray flower.
