%0 Journal Article %T 超高压协同温度处理对过敏原Bos d 6抗原性及二级结构的影响 %A 朱业培 %A 王玮 %A 吕青?V %A 滕爽 %A 徐幸莲 %A 周光宏 %J 南京农业大学学报 %D 2016 %R 10.7685/jnau.201509005 %X [目的] 本文旨在研究超高压协同温度处理对过敏原Bos d 6抗原性和二级结构的影响。[方法] 采用间接竞争ELISA和圆二色光谱方法,研究在25、40和55℃条件下,100、200、300、400、500和600 MPa压力分别处理10 min对过敏原Bos d 6抗原性及二级结构的影响,并分析过敏原抗原性与其二级结构中α-螺旋的相关性。[结果] 超高压处理能有效降低过敏原Bos d 6的抗原性及其二级结构中α-螺旋含量;随着处理温度的增加,过敏原抗原性和α-螺旋含量的降低幅度明显;超高压协同温度处理后过敏原Bos d 6抗原性与α-螺旋含量呈明显的正相关性。[结论] 超高压协同温度处理技术可以通过改变过敏原Bos d 6蛋白质的空间构象来达到降低其抗原性的目的。</br>[Objectives] The effect of combined ultra high pressure and temperature treatment on the allergenicity and secondary structure of allergen Bos d 6 was investigated.[Methods] A study was carried out about the ultra high pressure(100,200,300,400,500 and 600 MPa)for 10 min at 25,40,and 55℃ on the allergenicity and secondary structure of allergen Bos d 6 using indirect competitive ELISA and circular dichroism spectroscopy. The correlation between the α-helix content and the allergenicity was analyzed by linear regression.[Results] The allergenicity and α-helix content in the secondary structure of allergen Bos d 6 could be effectively reduced by ultra high pressure,which decreased significantly with the temperature increasing. There was a significant positive correlation between the allergenicity and α-helix content of allergen Bos d 6 after the combined treatment of ultra high pressure and temperature treatment.[Conclusions] The combined ultra high pressure and temperature treatment could decrease the allergenicity of allergen Bos d 6 through changing their protein conformation %K 超高压 %K 温度 %K 过敏原Bos d 6 %K 抗原性 %K 二级结构< %K /br> %K ultra high pressure %K temperature %K allergen Bos d 6 %K allergenicity %K secondary structure %U http://nauxb.njau.edu.cn/oa/darticle.aspx?type=view&id=201604020
