%0 Journal Article %T 高产与低产蛋鸡卵巢和卵泡及相关基因的表达差异 %A 李佳宜 %A 陆应林 %A 刘小凡 %A 虞德兵 %J 南京农业大学学报 %D 2017 %R 10.7685/jnau.201611031 %X [目的]以东乡绿壳蛋鸡为研究对象,从形态学和分子水平上探究高低产蛋鸡卵巢和卵泡的差异。[方法]选择高产组、低产组东乡绿壳蛋鸡各10只,采集静脉血,测定相关激素含量;屠宰采集卵巢组织,制作组织切片,观察卵巢形态结构;提取卵巢组织RNA,分别测定高产与低产蛋鸡卵巢中的细胞增殖凋亡基因、生殖调控相关转录因子基因、类固醇激素合成关键酶基因和相关激素受体基因的表达量。[结果]高产与低产蛋鸡卵巢和卵泡在形态上有明显差别:高产蛋鸡卵巢体积大,卵泡丰富,细胞排列紧密;低产蛋鸡卵巢、卵泡发育不良,细胞松散。高产蛋鸡血清中促卵泡激素(FSH)、孕酮和雌二醇水平均显著高于低产蛋鸡(P<0.05)。实时定量PCR检测结果显示:在高产蛋鸡中翼状螺旋/叉头转录因子2(FOXL2)、转录因子GATA家族4基因(GATA4)和B淋巴细胞瘤2基因(Bcl2)表达显著上调(P<0.05),且在高产蛋鸡卵巢中激活素受体1(ACVR1)和增殖细胞核抗原基因(PCNA)表达量极显著高于低产蛋鸡(P<0.01),但细胞色素P450家族11A1基因(CYP11A1)表达量显著低于低产组(P<0.05)。[结论]血清中生殖激素含量以及FOXL2、GATA4、Bcl2、ACVR1、PCNA和CYP11A1基因表达的差异是影响鸡产蛋性能高低的重要因素。</br>[Objectives]Dongxiang blue-shelled-egg chickens were used as the experiment object to explore the difference of ovary and follicle between high-and low-laying hens from morphology and molecular level,and provide theoretical basis for further study of genes involved in the regulatory mechanism of follicle development and follicle-microenvironment.[Methods]Ten high-laying hens and 10 low-laying hens were chosen as samples,venous blood were collected to determine the amounts of hormones. Ovary tissues were collected after slaughter and tissues were made into tissue slices to observe the morphological structure of ovary. RNA were extracted from ovary tissues to determine the gene expression of cell proliferation and apoptosis,transcription factors related to reproductive regulation,key enzyme of steroid hormone synthesis and their relative receptors in ovary.[Results]The results indicated that there were significant differences between high-and low-laying hens in morphology of ovary and follicle. High-laying hens had larger ovary,more follicle and cells arranged more closely than low-laying hens. The levels of follicle-stimulating hormone(FSH),progesterone(P)and estradiol(E2)were all higher in high-laying hens than that in low-laying hens(P<0.05). The result of real time PCR of all genes showed that FOXL2,GATA4 and Bcl2 expressions in high-laying hens were significantly higher than those in low-laying hens(P<0.05). And the expressions of ACVR1 and PCNA in high-laying ovary were significantly higher than those in low-laying hens ovary.
