%0 Journal Article %T HPLC-ELSD测定灵芝及其孢子粉中胆碱质量分数 %A 王亚涛 %A 刘艳芳 %A 张芝华 %A 张忠 %A 张劲松 %A 王晨光 %A 周帅 %A 唐庆九 %J 食品与生物技术学报 %D 2018 %R 10.3969/j.issn.1673-1689.2018.01.006 %X 建立了一种运用高效液相色谱-蒸发光散射检测器(HPLC-ELSD)测定灵芝及其孢子粉中胆碱含量的方法。实验采用Agilent公司ZORBAX 300-SCX阳离子交换色谱柱(4.6 mm×250 mm,5 μm),柱温30 ℃,流动相为乙腈-乙酸铵溶液,等度洗脱,流量为1.0 mL/min,进样量为10 μL;蒸发光散射检测器检测,漂移管温度为60 ℃,喷雾器为模式加热,雾化气为净化空气,气体压力为172.4 kPa。研究结果表明,该方法精密度、重复性、稳定性和回收率均良好,在6.25~200 μg/mL范围内胆碱具有良好的线性关系,检出限为6 μg/mL,定量限为12 μg/mL,是一种快速高效检测灵芝及其孢子粉中胆碱的方法。</br>To establish an HPLC-ELSD method for determining the content of choline in Ganoderma lucidum spore powder. The experiment using Agilent company ZORBAX 300-SCX cation exchange chromatography column(4.6 mm×250 mm,5 μm)as the stationary phase,the column temperature was set at 30 ℃,the mobile phase was 0.3% acetonitrile - ammonium acetate solution(60∶40),with isocratic elution,the flow rate was 1.0 mL/min,the injection volume was 10 μL,the detector was evaporative light -scattering detector(ELSD),the drift tube temperature of the ELSD was set at 60 ℃,the spray mode was heated,and the purifying air pressure was 172.4 kPa. Research results show that the method precision,repeatability,stability and recovery rate are good.The choline showed a good linear relationship in the range of 6.25~200 μg/mL.
