%0 Journal Article %T 基于巨噬细胞模型的竹荪多糖的免疫功能 %A 尚京迎 %A 付海田 %A 邓超 %A 张斯秀 %A 陈敬华 %J 食品与生物技术学报 %D 2016 %R 10.3969/j.issn.1673-1689.2016.08.010 %X 为探索竹荪多糖(DI)对巨噬细胞RAW264.7免疫功能的影响,分别采用四唑氮蓝(MTT)法、中性红法、Griess法、ELISA法,检测DI对巨噬细胞增殖、吞噬活性、一氧化氮(NO)和白细胞介素1(IL?鄄1)、白细胞介素6(IL?鄄6)、α肿瘤坏死因子(TNF?鄄α)分泌量的影响;运用反转录PCR(RT?鄄PCR)检测iNOS和IL?鄄1、IL?鄄6、TNF?鄄α的mRNA表达量的变化。结果表明,在25~200 μg/mL质量浓度范围内,DI能够明显促进巨噬细胞增殖,增强其吞噬活性。同时,DI能够增强巨噬细胞分泌NO、IL?鄄1、IL?鄄6、TNF?鄄α的能力,且明显提高iNOS、IL?鄄1、IL?鄄6、TNF?鄄α mRNA的表达水平。因此,竹荪多糖对巨噬细胞RAW264.7具有免疫刺激作用。</br>In order to investigate the immunomodulatory effects of a polysaccharide from Dictyophora indusiata (DI) on the immune function of RAW264.7 cells,the effects of DI on cell proliferation,phagocytosis of RAW264.7 cells,levels of NO,IL-1,IL-6 and TNF-α production were detected by MTT assay,neutral red test,Griess method,and ELISA,respectively. The mRNA expression level of iNOS,IL-1,IL-6 and TNF-α were also measured by reverse transcription polymerase chain reaction (RT-PCR). The results indicated that DI contributed significantly to the proliferation of the macrophages and the phagocytic activity was at 25~200 μg/mL. In addition,the production of NO,IL-1,IL-6 and TNF-α in macrophages were increased. Further,the expression of iNOS,IL-1,IL-6 and TNF-α mRNA were significantly up-regulated.
