%0 Journal Article %T 磷酸甘油酸变位酶1在肿瘤发生及发展中的作用<br>Role of phosphoglycerate mutase 1 in the occurence and development of tumor %A 梁倩 %A %A 沈瑛 %A < %A br> %A LIANG Qian %A %A SHEN Ying %J 上海交通大学学报(医学版) %D 2018 %R 10.3969/j.issn.1674-8115.2018.04.021 %X 磷酸甘油酸变位酶1(phosphoglycerate mutase 1,PGAM1)作为糖酵解通路的重要酶之一,在多种肿瘤组织中高表达,并且与肿瘤患者预后呈负相关。PGAM1催化糖酵解通路中3-磷酸甘油酸(3-phosphoglycerate,3-PG)转化生成2-磷酸甘油酸(2-phosphoglycerate,2-PG),促进葡萄糖代谢和能量生成。PGAM1通过调节3-PG与2-PG的转化平衡来影响其他代谢通路,参与细胞内生物大分子合成和维持氧化还原稳态,促进肿瘤细胞增殖及转移。PGAM1作为潜在的抗肿瘤靶标,其抑制剂的研发也成为抗肿瘤药物研究热点之一。该文对PGAM1结构、调控、在肿瘤发生及发展中的作用以及PGAM1抑制剂的最新研究进展作一综述。<br>:Phosphoglycerate mutase 1 (PGAM1) as one of the most important enzymes for glycolysis pathway, is highly expressed in multipletumor tissues and negatively correlated with the prognosis of cancer patients. PGAM1 catalyzes the conversion of 3-phosphoglycerate (3-PG) to2-phosphoglycerate (2-PG) in glycolysis pathway, then promoting anabolic pathways, energy generation, and maintaining redox balance during cancercell proliferation and metastasis. The small molecule PGAM1 inhibitors have emerged as a promising strategy for anti-tumor therapy.
