%0 Journal Article %T 毛果杨HDAC基因家族序列及其表达分析<br>Sequence and gene expression of histone deacetylases (HDAC) gene family of Populus trichocarpa %A 李淑娟 %A 张 %A 超 %A 张彦妮 %A 等 %J 西北农林科技大学学报(自然科学中文版) %D 2015 %X 【目的】研究毛果杨组蛋白去乙酰化酶(HDAC)基因家族序列及其对脱落酸(ABA)的应答反应,为HDAC家族基因的功能研究奠定基础。【方法】采用生物信息学方法对毛果杨HDAC家族的16种蛋白进行系统分析;采用实时荧光定量 PCR方法,分析了100 μmol/L ABA叶面喷洒处理后6和24 h毛果杨叶片HDAC家族16个基因的表达情况。【结果】HDAC家族蛋白分析显示,毛果杨HDAC可分为3个亚家族,即RPD3/HDA1、HD2和SIR2亚家族,与拟南芥HDAC蛋白的亲缘关系较近。HDAC家族大多数成员为亲水性蛋白质(HDA912属于疏水性蛋白),其等电点呈酸性(除了HDA912和SIR2亚家族)。HDAC家族蛋白受磷酸化调节,磷酸化位点主要发生在丝氨酸残基上。绝大部分HDAC蛋白都有不同数目的跨膜螺旋和不同的跨膜方向,而在HD2蛋白中没有发现跨膜区。二级结构分析显示,RPD3/HDA1和SIR2亚家族成员均含有不同比例的α-螺旋、β-折叠片和无规则卷曲,其中无规则卷曲比例达到50%以上(HDA912除外);而HD2亚家族成员不含有α-螺旋,其二级结构主要以无规则卷曲为主,比例高达80%左右。HDAC家族蛋白主要定位于细胞核内,在细胞质、细胞器及其他膜结构中也有分布,HD2蛋白几乎都定位于细胞核内。RPD3/HDA1和SIR2亚家族成员的三级结构可分为6种类型,预示其生物学功能也可能存在差异。实时荧光定量 PCR分析结果表明,ABA处理6 h显著提高了毛果杨-HDA901、HDA903和HDT-901基因的表达,而ABA处理24 h则会显著降低-HDA907和HDT-901基因的表达。【结论】作为植物特有的一类组蛋白去乙酰化酶,毛果杨HD2亚家族蛋白在序列和结构等多个方面与其他2个亚家族不同。ABA能够调节杨树HDAC家族基因的表达,HDAC家族基因不同成员对ABA的应答反应不同。<br>【Objective】The sequence and expression of histone deacetylase (HDAC) family genes of Populus trichocarpa and their response to ABA were investigated to provide useful information for future functional studies.【Method】16 HDAC proteins from Populus trichocarpa were analyzed with the bio-informatics tools.The expression levels of the 16 HDAC family genes in leaves after 100 μmol/L ABA treatment for 6 and 12 h were analyzed using real-time PCR.【Result】The populus HDACs could be classified into three sub-families,including RPD3/HDA1,HD2 and SIR2,and they had close relationship with HDACs from Arabidopsis thaliana.Almost all of the HDACs (except HDA912) were hydrophilic proteins and their isoelectric points (pI) were low (except for HDA912 and SIR2 sub-family).HDACs activities were regulated by phosphorylation,whichmajorly occured at the serine residues.Most of the populus HDACs had trans-membrane helices with different numbers and directions,whereas no helices were predicted in HD2 proteins.For RPD3/HDA1 and SIR2 proteins,the secondary structures included α-helices,β-strands and loops,and the percentages of loops were as high as 50% (except for HDA912).The three populus HD2 proteins had no α-helices in their secondary structures,and the loops were their primary secondary structure,accounting for ~80%.Populus HDACs were majorly localized in nucleus.They also distributed in organelles,cytoplasm or membrane components,whereas HD2 proteins were exclusively localized in nucleus.The tertiary structures of populus HDACs could be classified into 6 types,suggesting that they may have different functions.Real-time PCR analysis showed that the expression of HDA901,HDA903 and HDT901 genes were significantly improved by 6 h ABA treatment,whereas the expression of HDA907 and %K 毛果杨 %K 组蛋白去乙酰化酶 %K 生物信息学 %K 脱落酸(ABA) %K 基因表达< %K br> %K Populus trichocarpa %K histone deacetylase %K bio-informatics %K abscisic acid (ABA) %K gene expression %U http://www.xnxbz.net/xbnlkjdxzr/ch/reader/view_abstract.aspx?file_no=20150310&flag=1
