%0 Journal Article %T NDV、KLT及其联用时小鼠H22肝癌瘤组织的病理变化与TNF-α表达的研究<br>Effects of NDV,KLT and their combination on pathological changes of tumor tissue and expression of TNF-α of mice with H22 liver cancer %A 周 %A 琳 %A 张 %A 琪 %A 梁 %A 帅 %A 等 %J 西北农林科技大学学报(自然科学中文版) %D 2015 %X 【目的】研究新城疫病毒(New Castle Disease Virus,NDV)、康莱特注射液 (Kanglaite Injection,KLT)及NDV+KLT联用治疗肝癌肿瘤过程中,癌组织的病理变化以及肿瘤坏死因子-α(Tumor necrosis factor-alpha,TNF-α)在肿瘤组织中的表达情况,探究TNF-α在抗肿瘤过程中的作用,为应用生物素治疗肿瘤提供一定的理论依据。【方法】建立小鼠H22肝癌肿瘤模型,将96只荷瘤小鼠随机均分成生理盐水对照组(0.8 mL/只)、NDV治疗组(0.8 mL/只)、KLT治疗组(0.5 mL/只)及NDV+KLT联合治疗组共4组,分别在治疗后的4,8,12和16 d取小鼠肿瘤组织制作2套石蜡切片,一套进行HE染色检测病理变化;另一套进行免疫组织化学SP观察TNF-α的表达情况。【结果】HE染色发现,与对照组相比, NDV组和KLT组都有明显的肿瘤生长抑制及肿瘤细胞坏死现象,特别是NDV+KLT联合治疗组的治疗效果显著;免疫组化研究结果表明,不同的治疗组TNF-α分布差异显著,其中NDV+KLT联合治疗组的阳性反应最强,其次是KLT治疗组。【结论】NDV和KLT的抗肿瘤作用与TNF-α细胞因子表达有一定的相关性,其中NDV+KLT联合治疗的抗肿瘤效果最好。<br>【Objective】This study used New Castle Disease Virus (NDV),Kanglaite Injection (KLT) and their combination to treat liver cancer.The pathological changes of tumor tissue and expression of tumor necrosis factor-alpha (TNF-α) were analyzed to understand the function of TNF-α in cancer treatment.【Method】Mice H22 tumor model was established with 96 tumor-bearing mice randomly divided into four groups,NS control group (0.8 mL),NDV treatment group (0.8 mL),KLT treatment group (0.5 mL) and NDV+KLT treatment group.Tumor tissue was obtained from every group in the 4,8,12 and 16 d respectively.Paraffin sections were made to observe pathological changes using hematoxylin-eosin (HE) staining and expression of TNF-α using immunohistochemistry SP method.【Result】HE staining showed that,compared with NS control group,tumor growth inhibition and tumor cells necrosis appeared,especially NDV+KLT treatment group had significant treatment effect.Compared with NS control group,pathological changes of tumor tissue and distribution of TNF-α were significantly different in all treatment groups,especially in NDV+KLT group.NDV+KLT group also had the strongest positive reaction,followed by KLT treatment group.【Conclusion】Anti-tumor effect of NDV and KLT was related to expression of TNF-α and combination of NDV and KLT had the best effect %K TNF-α %K NDV %K KLT %K H22肝癌< %K br> %K TNF-α %K NDV %K KLT %K H22 liver cancer %U http://www.xnxbz.net/xbnlkjdxzr/ch/reader/view_abstract.aspx?file_no=20150703&flag=1
